摘要
[目的]寻找一条从植物中提取出较纯净SOD的廉价工艺路线。[方法]利用超氧化物歧化酶(SOD)耐热等性质,对以猕猴桃为原料制备的SOD粗酶液分别进行热变性、等电点、丙酮沉淀处理,以得到纯度较高的SOD酶液。[结果]试验确定了热变性、等电点、丙酮沉淀处理猕猴桃中的SOD粗酶液的最佳提取参数分别为65℃下25 min、pH 5.0、V丙酮∶V SOD酶液=1∶1,并且依次用3种方法下的最佳参数进行提取,得到的蛋白样品经分析达到了电泳纯。[结论]研究可为植物中SOD的廉价提取提供参考依据。
[Objective] To finda cheap technique route forextracting SOD from plants. [Method] Taking advantage of the heat-resistant nature and some other natures of superoxide dismutase, the crude enzyme of SOD from kiwifruits which were the raw material, was processed under the conditions of thermal denaturation, isoelectric point and precipitation by acetone. [ Result ] and these optimal extraction parameters were respec-tively determined as 25 min under 65 ℃, pH = 5.0, Vaeetone : VSODentyme= 1 : 1. Moreover, these optimal extraction parameters under the condi-tions were in turn used for extraction, and the sample of protein extracted reached the purity of electrophoresis. [ Conclusion ] A simple and cheap extraction process route of SOD was determined.
