摘要
观察抗癫痫中药草果知母汤在阻断戊四唑点燃模型形成过程中 ,对兴奋性氨基酸类神经递质Glu的受体NMDA结合力及其NMDAR1mRNA的影响。以NMDA的竞争性拮抗剂MK 80 1为对照 ,用配基受体法、原位杂交法检测相应指标的变化。结果显示 :与模型组相比 ,中药组大鼠海马NMDAR1mRNA的含量明显降低 ,差别有显著性意义 (P <0 0 5) ,MK 80 1组无明显变化 ;与模型组相比 ,中药组NMDA受体结合力明显降低 ,但差别无显著性意义 ,MK 80 1组皮层NMDA结合力明显降低 ,差别有极显著性意义 (P <0 0 1)。说明中药草果知母汤主要通过降低NMDAR1mRNA的表达来降低NMDA受体的含量。
The effects of Caoguozhimu Decoction (CD), a TCM antiepileptic, on NMDA receptor binding capacity and the expression of NMDAR1 mRNA in pentylenetetrazole kindled rat model were observerd by the ligand receptor method and in situ hybridization, with MK 801 as a contrast. The results showed that compared with model group, the level of hippocampal NMDAR1 mRNA was significantly decreased ( P <0 05) in CD group, while there was no obvious change of that in MK 801 group; and the cortical NMDA receptor binding capacity was obviously decreased but without statistical significance in CD group, while there was a significant decrease ( P <0 05) of that in MK 801 group. The results imply that CD decreases the level of NMDA receptor mainly by decreasing the expression of NMDAR1 mRNA.
