摘要 目的介绍一种改良的单细胞凝胶电泳技术同时检测血白细胞DNA链断裂和DNA-蛋白交联并将其应用于部分健康人群。方法60名健康人群,每人血样分A、B两份,B份在细胞裂解后加蛋白酶K,其余同常规方法;B份尾矩为总的DNA链断裂量,B份与A份尾矩的差值为DNA-蛋白交联量。结果DNA链断裂在性别、年龄和饮酒与否方面的分布无显著性差异,而吸烟可使其显著增加;上述各因素对DNA-蛋白交联均无显著影响,后者在该研究人群中的分布里正态(2.46 ×106± 0.70 ×106)。结论该研究为职业接触人群 DNA损伤的监测提供了方法学和对照数据的参考。 Objective To introduce a modified single cell gel electrophoresis for detecting DNA strand breaks (DSB) and DNA-protein crosslinks (DPC) in white blood cells simultaneously, and apply it to healthy population. Methods Each blood sample from 60 healthy people was divided into two shares (A and B), adding proteinase K into B after lysis. The other procedures were the same as ordinary method. Tailmom in B was foe total content of DSB and the different tailmom between B and A was that of DPC. Results it seemed to have no significant differences in DSB among sex, age and alcohol drinking, but it did due to smoking. All the factors have no marked influence on DPC in healthy population as it was in normal distribution (2 .46 ×106± 0 .70 ×106). Conclusion This study offered references in methodology and control data for monitoring DNA damage in occupational population.
