转基因大豆外源基因CaMV35S启动子测定不确定度分析 被引量:4

摘要 为了了解转基因成分定量分析结果不确定度的主要贡献因素,提高转基因成分定量检测质量,本文对转基因大豆GTS40-3-2粉末样品进行了CaMV35S启动子(参数)含量测定,对其测量不确定度进行了初步估算。CaMV35S启动子测量不确定度评定结果:A类不确定度(uA)为0.0004,B类不确定度(uB)为0.002,合成不确定度(uc)为0.002;在置信水准(p)95%时,包含因子(k)取1.96,扩展不确定度(U)为0.004。本次测量结果(C)为0.028±0.004,接近约定真值(0.03),测量不确定度相对较小,检测质量较高。不确定度评估表明试验中的微量液体转移偏差是本次测量不确定度的主要贡献者,在以后的检测中应重点注意降低微量液体转移的偏差,提高检测质量。 A preliminary assessment of measurement uncertainty of CaMV35S promotor detection in transgenic soybean samplea was made in this paper to understand the knowledges of the main contributors among the sources of uncertainty and to further increase the quality of testing in future experiments.The uncertainty estimates:uncertainty (uA) of type A was 0.0004,type B was (us) 0.002,and the combined uncertainty (uc was 0.002; When confidence level (P) was 95 % and the coverage factor (k) was 1.96,then,expanded uncertainty (U)was 0.004.Consequently,this measurement result(C) was 0.028 ± 0.004,close to the conventional true value (0.03),and the measurement uncertainty is relatively small,reflecting a high quality of detection.As a result,the trace amount of liquid transfer was the major source of measurement uncertainty in this experiment.Thus,the trace amount of liquid transfer should be noted in future detection.
