暗双孢菌AGR0073产生的单端孢菌素的分离纯化、结构解析及其抗植物病原真菌活性 被引量:2

摘要 在开展云南山地香蕉根际和内生真菌分离及其次生代谢产物生物活性筛选过程中,获得一株对香蕉枯萎病菌具有强拮抗作用的真菌(Cordana musae,AGR0073)。分别用乙酸乙酯和石油醚对菌株发酵液进行活性物质的提取分离,并分别通过正相硅胶柱层析、反相C18硅胶柱层析、凝胶(Sephadex LH-20)柱层析和制备HPLC纯化,从菌株发酵液中分离获得具有抗真菌活性的化合物FM 9-1。通过1H-NMR和13C-NMR波谱分析以及结合相关文献对比,将该化合物鉴定为单端孢菌素(Trichothecin,TCN),这是首次从Cordana musae代谢产物中分离出该化合物。抑菌活性测定表明,化合物FM 9-1具有抗植物病原真菌活性,对香蕉枯萎病菌(Fusarium oxysporum f.sp.cubense)、香蕉炭疽病菌(Colletotrichum musae)、香蕉小窦氏叶斑病菌(Deightoniella torulosa)、香蕉弯孢霉叶斑病菌(Curvularia lunata)、香蕉角斑梨孢病菌(Pyricularia angulata)等5个病原真菌最低抑制浓度(Minimum inhibitory concentration,MIC)分别为1.4、1.1、6.5、2.3和5.2μg/mL。 Endophytic and rhizosphere fungi from plants were important sources for microorganisms which could produce a variety of bioactive compounds.In this study,a fungus Cordana musase AGR0073 with high antifungal activity was isolated from banana in Yunnan mountain plateau bananas.An antifungal compound FM 9-1 was isolated and purified from the fermentation hoth of AGR0073 through extraction with ethyl acetate and petroleum ether,normal-phase silica gel column chromatography,reversed-phase Cn silica gel column clromatography,LH-20 gel column chromatography and preparative HPLC,traced by ultraviolet light or iodine vapor with biossay-guided fractionation.FM 9-1 was elucidated as Trichothecin(TCN) based on mass spectrometry,nuclear magnetic resonance spectroroopy as well as other relevant literature.The antifungal activity assays showed that compound FM 9-1 had strong sntlfungsl activity against Fusarium oxysporum L sp.cubense,Collewtrichum musae,Ddghtoniella tondosa,Cumdada lunata,Magnaponhe griseaa,and the minimum inhibitory concentrations were 1.4,1.1,6.5,2.3 and 5.2 μg/mL,respecvely.
