摘要
目的:观察扶正除疫颗粒对A型H1N1流感病毒感染小鼠T细胞亚群的影响,探讨其抗A型H1N1流感病毒感染的作用特点和机制。方法:40只BALB/c小鼠随机分为正常对照组、病毒模型组、达菲对照组、扶正除疫组。正常对照组和病毒模型组以0.9%氯化钠溶液灌胃,另两组以达菲、扶正除疫颗粒水溶液灌胃,连续7d。除正常对照组外,其余各组于给药第3天,以FM1-6-E2滴鼻造模。每组于造模后3、6、9d分别取脾脏制备细胞悬液,采用流式细胞仪测定T细胞亚群的变化。结果:各组与同期病毒模型组相比,除第3天的CD3+T、第6天的CD4+T及第9天CD3+T和扶正除疫组CD4+T和百分率数值没有显著差异外,其余各组各时段的CD3+T、CD4+T、CD8+T及CD4+T/CD8+T百分率数值均有显著差异(P<0.01,P<0.05)。结论:扶正除疫颗粒对A型H1N1流感病毒感染引起的细胞免疫功能失常有一定的调节作用。提示该药物抗流感病毒的作用机制,可能是通过调整T细胞亚群的百分率及其CD4+T/CD8+T比值得以实现。
Objective: To observe the effects of Fuzheng Chuyi Granule against H1N1 influenza A virus infection in murine T cell subset, and explore the characteristics and mechanism of action of Fuzheng Chuyi Granule in anti-influenza A virus infection. Methods: 40 BALB/c mice were divided into four groups at random, including the normal group, virus group, DF group and Fuzheng Chuyi group, with 10 mice in each group. 0.9% sodiurn chloride solution for the normal group and virus group by gavage, 0.4mL per mouse at one time; Dafei aqueous solution for DF group by gavage; Aqueous solution of Fuzheng Chuyi Granule for FZ group by gavage, seven days in total. On the third day of gavage, all the groups except the normal group, were established as the nasal model by use influenza virus FM1-6-E2. Selecting 3 mice from each group to be executed after three days, six days, and nine days of toxieation. Adopting flow cytometric assay to measure the change of T cell subset. Results: Compared with the virus group, except for the CD3+T on the 3rd day, the CD4+T on the 6th day, the CD3+T and CD4+T percentage values on the 9th day without significantly difference, segments of CD3+T, CD2+T, CD8+T and CD4+T/CD8+T percentage values of other groups on each time were significantly different (P〈0.01, P〈0.05). Conclusion: Fuzheng Chuyi Granule plays a regulatory role in reducing cellular immune function caused by the influenza virus, reducing the immune injury of the mice, to some extent. It is presented that the function mechanism of anti-flu virus of this kind of medicine might be fulfilled by adjusting to the percentage of CD3+T, CD4+T, CD8+T of T cell subtype and the ratio of CD2T/CD4+T.
