在EBV阳性鼻咽癌细胞内SAHA联合GCV细胞毒作用的时间点分析

摘要 目的探讨伏立诺他(SAHA)联合GCV对EBV阳性鼻咽癌细胞的细胞毒作用有效时间点,为临床化疗提供依据。方法利用Real-time PCR检测SAHA作用下EBV阳性鼻咽癌细胞C666 TK的表达情况;通过MTT检测SAHA联合GCV的细胞毒性。结果 SAHA作用16 h时TK的表达最多,并且MTT结果显示SAHA作用16 h后联合GCV两者的细胞毒作用最强。结论 SAHA作用16 h后联合GCV抗鼻咽癌效果较好。 Objective To explored the cytotoxicity action time point of SAI4A and GCV in Epstein-Barr virus (EBV) positive nasopharyngeal carcinoma (NPC) cells, so as to provide theory evidence tor climcal chemotherapy, metnoos EBV thymidine kinase (TK) expression was detected by real-time PCR in EBV positive NPC cells C666 with SAHA treatment. The cytotoxicity of SAHA and GCV was detected by MTT. Results The TK expression was most when SAHA treatment for 16 h, and the cytotoxicity effect of SAHA and GCV was the strongest at the same time. Conclusion The antitumor activities of SAHA and GCV in NPC cells is the best at SAHA treatment for 16 h.
