人甲状腺未分化癌鸡胚尿囊膜移植瘤模型的建立及生物学性状 被引量:1

Establishment of a transplantation tumor model of human anaplastic thyroid carcinoma in chick embryo and its biological characteristics

摘要 目的 建立人甲状腺未分化癌鸡胚尿囊膜(chick embryo chorioallantoic membrane,CAM)移植瘤模型,观察移植瘤的生长情况、组织形态及其与人甲状腺未分化癌组织形态的比较.方法收集本院2007~2011年11例甲状腺未分化癌患者的肿瘤组织,进行HE染色并分析;将一定量的甲状腺未分化癌细胞株FRO接种于CAM上,观察鸡胚的存活情况和移植瘤的生长情况、形态学特征及生物学特征,并取移植瘤组织进行HE染色.结果 20μL含有6×106的甲状腺未分化癌细胞株FRO可在鸡胚尿囊膜上生长且不影响鸡胚的活力,移植瘤形态学特征与人甲状腺未分化癌组织的形态学特征相似.结论 成功建立了人甲状腺未分化癌鸡胚尿囊膜移植瘤模型,为甲状腺未分化癌的进一步研究提供动物模型. Objective To establish transplantation tumor model of human anaplastic thyroid cancer in the chick embryo and observe the growth and morphology of transplantation tumor. Methods we collected tumor tissues of 1l patients with thyroid anaplastic carcinoma from 2007 to 2011 for HE staining and analysis. In a sterile environment, a certain amount of anaplastic thyroid cancer cell lines FROwas inoculated into chick embryo chorioallantoic membrane (CAM) to observe the survival of chick embryo as well as the growth, morphological characteristics and biological characteristics of transplantation tumor. Then the transplanted tumor tissue was taken for HE staining. Results Anaplastic thyroid cancer cell lines FRO of 20 μL containing 6 × 106 could grow in the chick embryo chorioallantoic membrane and did not affect the vitality of the chick embryo. The morphological characteristics of the transplantation tumor were similar to those of human thyroid undifferentiated cancer tissue. Conclusiorl We successfully constructed transplantation tumor model of human anaplastic thyroid cancer in the chick embryo. The model can provide an animal model for further study on anaplastic thyroid cancer.
