摘要
目的:采用反相高效液相色谱法分析武当山产葛花中葛花苷的含量。方法:采用LUNA—C18(250mm×4.6mm,5um)色谱柱,流动相为乙腈-0.1%磷酸溶液(18:82),流速为1.0mL/min,柱温30℃,检测波长265nm。结果:葛花苷在0.062~1.240mg/mL范围内与峰面积有良好的线性关系,平均加样回收率为99.1%,RSD为0.86%。结论:本文建立了葛花中葛花苷的反相高效液相色谱测定方法,为更全面评价武当山地区葛花药材质量提供依据,同时也为其他含有葛花苷成分的药材及成药分析提供了借鉴。
Objective: To establish the RP-HPLC method for the determination of kakkalide in Puerariae lobata Ohwio from Wudangshan. Methods: The LUNA-C18 column (250 mm×4.6 mm,5 um) was used; the mobile phase was consisted of acetonitrile- 0.1% phosphoric acid solution (18 : 82) ; the flow rate was 1.0 mL/min; the column temperature was 30 ℃ ; the detecting wavelength was set at 265 nm. Results: The calibration curve was linear within a range of 0. 062 - 1. 240 mg/mL; the average recovery of this method was 99.1%, and the RSD was 0.86%. Conclusions : This RP-HPLC determination method of kakkalide may provide basis for comprehensive evaluation of the quality of Puerariae lobata Ohwio in this region, and also provide references for ingredients analysis of other herbs containing kakkalide.
