摘要
目的 研究MAT1、nm2 3H1mRNA表达及突变与卵巢癌淋巴结转移的关系。 方法 应用逆转录 PCR(RT PCR)和逆转录 PCR 单链构像多态性分析 (RT PCR SSCP)技术 ,对 8例正常卵巢、2 0例卵巢癌及其相应的 2 0例淋巴结组织 ,进行MTA1和nm2 3H1mRNA表达及突变的检测。 结果 转移卵巢癌原发灶MTA1mRNA高表达率为 10 0 % (7/ 7) ,无转移为 38 5 % (5 / 13) ,P =0 0 10 3;有癌转移淋巴结高表达率为 87 5 % (6 / 7) ,无癌转移为 2 3 % (3/ 13) ,P =0 0 118。有转移卵巢癌原发灶nm2 3H1mRNA低表达率为 10 0 % (7/ 7) ,无转移为 30 % (4 / 13) ,P =0 0 0 43;有癌转移淋巴结低表达率为 10 0 %(7/ 7) ,无癌转移为 38 5 % (5 / 13) ,P =0 0 10 2。MTA1/nm2 3H1mRNA表达的相对吸光度值 (A值 ,曾称光密度OD值 )的比值随转移而增加。单链构像多态性分析 (SSCP)未发现突变。 结论 MTA1、nm2 3H1基因的转录表达与卵巢癌淋巴结转移呈正负相关关系 ,起着正负调控的重要作用。这两个基因的异常表达是卵巢癌转移中的频发事件而与基因突变无关。
Objective To investigate mRNA expression and mutation of MTA1 and nm23H_1 genes in ovarian carcinoma (OC) in relation to lymph node (LN) metastasis. Methods A panel of eight normal ovarian tissues, twenty primary OC specimens and twenty corresponding LNs was examined for mRNA expression and mutation of MTA1 and nm23H_1 genes by using RT-PCR and RT-PCR-SSCP. The level of expression was determined by the relative optic density (ROD) of the PCR products. [WT5”HZ]Results The frequency of MTA1 overexpression was 100% (7/7) in primary OC with metastasis but only 38.5% (5/13) in those without metastasis (P=0.010*!3). Overexpression of MTA1 was observed in 87.5% (6/7) of LNs with metastasis but in only 23% (3/13) of LNs without metastasis (P=0.011*!8). In contrast with MTA1, low expression of nm23H_1 mRNA was seen in 7 of 7 OC with metastasis but only in 4 of 13 (30%) of those without metastasis (P=0.004*!3). Low nm23H_1 expression was also seen in 7 of 7 LNs with metastasis but only in 5 of 13 (38.5%) nonmetastatic LNs (P=0.010*!2). The ROD ratio of MTA1 to nm23H_1 increased with the development of metastasis. No mutation of MTA1 and nm23H_1 was found by SSCP analysis. Conclusion[WT5”BZ] The mRNA expression of MTA1and nm23H_1 is positively and negatively correlated with LN metastasis, respectively. Expression abnormalities but not mutation of the two genes are frequent events related to LN metastasis of ovarian cancer.
