摘要
目的:研究附子干姜合用丙磺舒、兰索拉唑对一氧化氮合酶(NOS)抑制型高血压模型小鼠的影响,并初步探讨其作用机制。方法:将雄性ICR小鼠随机分为空白对照组(C组)、左旋N-硝基精氨酸(L-NNA)模型组(M组)、L-NNA+附子干姜组(FG组)、L-NNA+丙磺舒组(B组)、L-NNA+兰索拉唑组(L组)、L-NNA+附子干姜+丙磺舒组(FG+B组)、L-NNA+兰索拉唑+丙磺舒组(L+B组)、LNNA+附子干姜+兰索拉唑+丙磺舒组(FG+L+B组)。各组分别给予0.5%羧甲基纤维素钠(CMCNa)溶液、L-NNA、L-NNA+附子干姜水煎液3g/kg、L-NNA+丙磺舒360mg/kg、L-NNA+兰索拉唑30mg/kg、L-NNA+附子干姜水煎液3g/kg+丙磺舒360mg/kg、L-NNA+兰索拉唑组30mg/kg+丙磺舒组360mg/kg,L-NNA+附子干姜水煎液3g/kg+丙磺舒360mg/kg+兰索拉唑30mg/kg。持续灌胃给药10天后测定各组小鼠的血压,检测血清、心、肝、肾和胃组织中的一氧化氮(NO)和丙二醛(MDA)含量。结果:与C组相比,M组、B组、L组和L+B组血压显著升高(P<0.01,P<0.05,P<0.05),M组和B组大鼠的血清和心、肝、肾、胃中的NO含量下降,MDA含量升高(P<0.01或P<0.05)。与模型组相比,FG组、FG+B组、FG+B+L组小鼠的血压和生化指标均有所改善(P<0.01或P<0.05),其中FG+B组死亡率最低,FG+B+L组其次。结论:FG组、FG+B组、FG+B+L组药物均能升高NOS抑制型高血压小鼠的NO含量,降低MDA含量,从而产生降压、降低组织氧化损伤的作用,其中FG+B+L组小鼠更趋近于正常水平。
Objective:To study the effects and interactions of Radix Aconiti Lateralis Preparata combined with Rhizoma Zingiberis and probenecid, lansoprazole on nitric oxide synthase (NOS) inhibited mice. Methods:Male ICR mice were randomly divided into control group, model group, Radix Aconiti Lateralis Preparata combined with Rhizoma Zingiberis group (FG), probenecid group (B), lansoprazole group (L) , FG plus probenecid group (FG-bB), lansoprazole plus probenecid group (L+B) , FG plus probenecid and lansoprazole group (FG+L+ B). The mice were intragastricly administrated with CMC-Na, L-NNA, L-NNA-FFG, L-NNA+ B, L-NNA+L, L-NNA +FG+B, L-NNA+L+B, L NNA+FG+L+B, respectively for continuous 10 days. The changes of blood pressure were measured, as well as the contents of NO and MDA in serum, heart, liver, kidney and stomach. Results:Compared with control group, the blood pressure of M group, B group, L group and L+B group were increased significantly (P〈(0.01, P〈0.05, P〈0.05). The contents of NO in serum, heart, liver, kidney and stomach of M group and B group were sharply in- creased, and MDA decreased (P〈0.01or P〈0.05). Compared with the model group, FG group, FGq-B group and FG+L+B group were notably improved in blood pressure and biochemical indicators (P〈0.01or P〈0.05). The lowest mortality was among the mice in FG-LB group, and then the FG+L---B group. Conclusion: The FG group, FG+B group, and FG+B+L group respectively posed significant effect on the blood pressure, the contents of NO and MDA to protect the tissues from oxidative impairment, among which the mice in FG+B-FL group were closer to normal level.
