摘要
目的 研究吡拉西坦对原代培养海马神经元钙激活钾通道 (KCa通道 )的作用 ,以揭示吡拉西坦抗缺血性脑损伤的电生理机制。方法 应用膜片钳制技术记录原代培养的海马神经元的单通道电流活动 ,经 p- CL AMP软件进行微机采样储存数据和数据的分析处理。结果 吡拉西坦对 KCa通道具有明显的激活作用 ,增加通道开放概率 ,缩短通道关闭时间 (P<0 .0 1)。结论 吡拉西坦可能通过激活
Objective To investigate the effects of piracetam on Ca 2+ activated potassium channels in cultured newborn rat hippocampal neurons so as to reveal the electrophysiological mechanism about the effects of piracetam on the neurons of cerebral ischemia.[WT5”HZ]Methods Inside out method of patch clamp technique was adopted in this experiment to record the activity of single channel;p CLAMP software was used for data sampling,storing and analysing.Results After piracetam was applied to the cytoplasmic face of the channels,the effect of piracetam on Ca 2+ activated potassium channels was observed.It increased significantly the open probability and shortened the closed time of the channels( P <0.01).[WT5”HZ]Conclusion Piracetam may protect the ischemic neuron by activating the Ca 2+ activated potassium channels.
