摘要
目的分析不同浓度的水合氯醛对C57BL/6和KM两种雄性小鼠的麻醉效果。方法C57BL/6和KM两种雄性小鼠各60只,根据水合氯醛的浓度(2.5%、5%和10%)和剂量(a=0.06ml/10g和b=0.1 ml/10g)分为十二组,分别为C2.5a、C2.5b、C5a、C5b、C10a、C10b组和K2.5a、K2.5b、K5a、K5b、K10a、K10b组。水合氯醛经腹腔注射后,观察麻醉效果、诱导时间及维持时间。结果 KM小鼠随药物浓度增加,诱导时间逐渐缩短、维持时间渐长、麻醉渐深,而C57BL/6小鼠的C5b组诱导深麻时间最短、维持时间最长;C2.5a和K2.5a组仅有弱的镇静作用,而C10b和K10b组虽然诱导时间很短,但是小鼠均全部死亡。结论 C57BL/6和KM雄性小鼠手术造模的实验麻醉均建议按5%,0.1 ml/10g给药,短时的非手术造模C57BL/6小鼠可考虑以2.5%,0.1ml/10g给药,而KM小鼠则按5%,0.06ml/10g给药,可确保水合氯醛的麻醉安全性。
Objective To investigate the anesthetic effects of different chloral hydrate concentrations in two kinds of male mice; C57BL/6 and KM. Methods Depending on the concentration (2.5%, 5% and 10%) of chloral hydrate and doses (0.06 ml/10 g and 0.06 ml/10 g), mice were randomly divided into 12 groups, C2.5a, C2.5b, C5a, C5b, C10a, C10b and K2.5a, K2.5b, K5a, K5b, K10a, K10b,with 60 mice in each kind. After intraperitoneal injection of chloral hydrate, anesthetic effect, induction time and anesthesia maintenance time were recorded. Results With the increase of drug concentration in KM mice induction time was gradually shortened, the anesthesia maintenance time prolonged and depth of anesthesia gradually greater. But the induction time of C5b group in C57BL/6 mice was the shortest and the anesthesia maintenance time was the longest; C2.5a and K2.5a groups only had weak sedation effect. Although the induction time of C10b and K10b groups was very short, but the mice were all dead. Conclusion This operation mode of the experiment recommended to use 5%,0.1 ml/10 g for the two kinds of mice. 2.5%,0.1 ml/10 g for C57 BI./6 and 50/00,0.06 ml/10 g for KM are preferable in non-surgical short operation to make sure safety use of chloral hydrate anesthesia.
