Expression and Clinical Significance of Bone Morphogenetic Protein 6 in Human Breast Cancer

摘要 目的探讨骨形态发生蛋白6(bone morphogenetic proteins,BMP6)在乳腺癌组织的表达及与临床病理特征的关系。方法应用实时荧光定量RT-PCR方法检测BMP6mRNA在36例配对乳腺癌组织及癌旁组织中的表达情况;免疫组织化学染色方法检测BMP6蛋白在80例乳腺癌组织中的表达,并结合组织临床病理资料进行统计分析。结果实时荧光定量RT-PCR结果显示,BMP6mRNA在乳腺癌组织中的表达水平低于相对的癌旁组织(P<0.01)。BMP6mRNA在雌激素受体阳性(ER+)乳腺癌组织中的表达高于ER-乳腺癌组织,同时BMP6mRNA表达随乳腺肿瘤分化程度的增高而降低(P<0.01)。免疫组化染色结果显示,BMP6蛋白在不同ER、孕激素受体(progestogen receptor,PR)表达、增殖细胞核蛋白Ki-67表达指数及病理分级间的差异有统计学意义(P<0.05),在不同乳腺癌患者的年龄、肿瘤大小、人表皮因子受体2(Her2)表达状况以及乳腺癌分子亚型间的差异无统计学意义(P>0.05)。结论 BMP6分子在乳腺癌组织的低表达与乳腺癌的发生、发展有关,提示BMP6有可能成为鉴别乳腺良、恶性病变的重要分子指标,对乳腺癌的内分泌治疗等具有重要意义。 Objective To investigate the expression pattern and coincal significance of bone morphogeuetic protein 6 (BMP6) in breast tissues. Methods The tumor and adjacent noncancerous tissues were harvested from 36 cases of breast cancer, the expression level of BMP6 mRNA of each sample was measured by quantitative RT-PCR. lmmunohistochemistry study was used to examine BMP6 protein expression in 80 cases of breast cancer, then the relationship between the expression of BMP6 and relevant clinical and pathological parameters was analyzed. Results BMP6 mRNA expression in breast cancer was significantly reduced when compared with normal breast tissues (P〈 0. 01), BMP6 mRNA level in estrogen receptor-positive (ER+) breast cancer was distinctly higher than that in ER-breast cancer. The expression of BMP6 mRNA was correlated to tumor grade (P〈0.01). The expression level of BMP6 protein in breast cancer was associated to ER and PR status, histological grade and Ki-67 status (P〈 0.05), but not correlated to age, tumor size, human epidermal factor receptor-2 (Her2) status and molecular subtypes of breast cancer (P〈0.05). Conclusion The eetopic expression of BMP6 may play an important role in the development and progression of breast cancer.
