摘要
目的探讨原发性肝癌(PHC)患者血清miR-1的表达及其与临床病理特征之间的关系。方法采用实时定量PCR检测90例PHC患者及102例健康志愿者血清中miR-1的表达量。结果PHC组中miR-1表达量[0.210(0.127~0.312)]明显低于健康对照组[0.780(0.618~1.121)]及PHC术后7d组[0.540(0.318—0.716)](P〈0.001)。在90例PHC患者中,miR-1表达水平与淋巴结有无转移密切相关,有淋巴结转移患者中miR-1表达水平显著低于无淋巴结转移患者(P=0.003)。Kaplan—Meier分析显示miR-1低表达组的生存率远低于其高表达组,其中位生存期(月)分别为25.48和40.98(P〈0.001)。Cox多变量分析显示miR-1水平是影响PHC患者生存率的独立危险因素之一,相对危险度为0.424(95%CI:0.183—0.984)。结论血清miR-1作为新的生物学指标对于PHC的诊断和预后的具有一定的价值。
Objective To investigate the serum expression level of miR - 1 in patients with primary hepatic carcinoma (PHC) and its clinical significances. Methods The serum expression levels of miR - 1 in 102 healthy controls and 90 PHC patients were measured by real -time fluorescent quantitative -polymerase chain reaction (qRT- PCR). Results Serum miR - 1 ex- pression level in PHC pre - operative group[0. 210(0. 127 - 0. 312) ] was significantly lower than that in the control group [ 0. 780 (0.618 ± 1. 121 ) ] and the post - operative group [ 0.540 (0.318 ± 0.716 ) ] ( P 〈 0.001 ). Serum miR - 1 expression level was far lower in PHC patients with lymph node metastasis than those without, showing close correlation between miR - 1 expression level and lymph node metastasis ( P = 0.003 ). Kaplan - Meier curve analysis revealed that PHC patients with a low serum miR - I expression level exhibited significant short survival time when compared to those with a high serum miR - 1 ex- pression level (25.48 months vs 40.98 months, P 〈 0. 001 ). Multivariable Cox regression analysis indicated that the serum miR - 1 expression level was an independent risk factor for survival rate of PHC patients. Relative risk was 0.424 (95% confi- dence: 0. 183 - 0. 984). Conclusion As a new biological marker, serum miR - 1 is helpful to diagnose PHC and judge the progression and prognosis of the disease.
