叶黄颗粒对光损伤大鼠视网膜及血清游离氨基酸的影响

Effects of lutein granule on retina and serum free amino acid in rats with retina light impairment

摘要 目的观察叶黄颗粒对光损伤大鼠视网膜及血清游离氨基酸的影响。方法将48只大鼠随机分为6组,每组8只。建立光损伤模型,于光照后7 d,测定大鼠视网膜及血清游离氨基酸含量。结果光损伤后视网膜中谷氨酸(Glu)、天门冬氨酸(Asp)、甘氨酸(Gly)、γ-氨基丁酸(GABA)、牛磺酸(Tau)5种神经递质类游离氨基酸及苏氨酸(Thr)、缬氨酸(Val)、蛋氨酸(Met)、异亮氨酸(Ile)、亮氨酸(Leu)、苯丙氨酸(Phe)、赖氨酸(Lys)、组氨酸(His)、精氨酸(Arg)、丝氨酸(Ser)、丙氨酸(Ala)、瓜氨酸(Cit)、半胱氨酸(Cys)、酪氨酸(Tyr)及鸟氨酸(Orn)等其他15种必需和非必需游离氨基酸含量均升高(P<0.05或P<0.01);血清中的上述游离氨基酸含量未见明显升高(P<0.05)。结论叶黄颗粒可以有效抑制光损伤后视网膜游离氨基酸水平的增高,可能是其保护视网膜光损伤的机制之一。 Objective To observe the effect of lutein granule on retina and serum free amino acid in rats with retinal light dam- age. Methods Forty -eight rats were randomly divided into 6 groups, 8 rats in each group. The retinal light damage model was established, then the contents of free amino acids in rat retina and serum were determined after 7 - day light. Results M- ter the light damage, the contents of 5 neurotransmitter free amino acid, including glutamate (Glu), aspartic acid (Asp), gly- cine (Gly), gamma aminobutyric acid (GABA) and taurine (Taa), as well as that of other 15 kinds of essential and non -es- sential amino acids including threonine (Thr), valine (Val), methionine (Met), isoleucine (Ile), leucine(Leu), phenylala- nine (Phe), lysine (Lys), histidine (His), arginine (Arg), serine (Ser), alanine (Ala), citrulhne (Cit), cystenine ( Cys), tyrosine (Tyr) and ornithine ( Orn), increased significantly in retina ( P 〈 0.05 or P 〈 0.01 ) ; while significant in- crease was not found in above free amino acid contents in serum (P 〉 0.05). Conclusion Lutein granule can effectively inhibit the increase of free amino acid levels in retina after light damage, which may be a protective mechanism of retinal light damage.
