Optimization of Extraction Technique of Tobacco Stem Using Response Surface Methodology

摘要 利用响应面法对造纸法再造烟叶生产中烟梗萃取工艺进行优化。通过单因素实验考察了萃取温度、萃取时间和液料比对烟梗萃取率的影响。采用中心组合实验设计,建立以萃取率为响应值的响应面方程。在此基础上,建立了萃取效益优化模型,模型综合考虑了萃取率、萃取成本与效率。应用响应面法对模型进行分析。所得烟梗萃取的最佳工艺条件为:萃取温度70℃,萃取时间50 min,液料比8.3,此时萃取率为31.81%,萃取效益最高。 Response surface method (RSM) was used to optimize the extraction condition of the tobacco stem on producing papermaking-reconstituted tobacco. The effects of extraction temperature, extraction time and ratio of solvent to raw material on extraction rate of tobacco stem were investigated by single factor tests. Base on the single factor tests, an equation was established with CCD (central composite design). On the basis of this equation, a extraction benefit model which evaluates the effects of extraction rate, cost and efficiency was established. RSM was applied to optimize this model. The result showed that the optimum conditions for the tobacco stem extraction were as following: the extraction temperature 70℃, the extraction time 50 min and ratio of solvent to raw material 8.3 mL·g^-1. Under the optimum conditions, the extraction rate is 31.8% and the extraction benefit is the best.
