人参皂苷Rg_3对失巢凋亡耐受乳腺癌细胞MCF-7抑制作用 被引量:10

摘要 目的研究人参皂苷Rg3对失巢凋亡耐受的乳腺癌细胞MCF-7生长抑制作用。方法应用多聚2-羟乙基甲基丙烯酸酯(poly-hema)覆盖的细胞培养板,筛选出具有失巢凋亡耐受的乳腺癌细胞MCF-7,考察失巢凋亡耐受癌细胞的生长速率和侵袭力,并利用流式细胞仪检测人参皂苷Rg3对失巢凋亡耐受癌细胞MCF-7的周期分布和细胞凋亡。结果与阿霉素对照组相比,人参皂苷Rg3能显著抑制失巢凋亡耐受的乳腺癌细胞MCF-7的生长(P<0.01),并导致失巢凋亡耐受细胞MCF-7的G0/G1期细胞比例显著增加(P<0.05)。结论人参皂苷Rg3有效抑制失巢凋亡耐受的乳腺癌细胞MCF-7生长,其作用机制可能与人参皂苷Rg3使失巢凋亡耐受乳腺癌细胞MCF-7细胞周期停滞有关。 Objective To investigate the effect of ginsenoside Rg3 on the cell growth and the cell cycle arrest in anoikis-resistant breast cancer cell MCF-7. Methods The anoikis-resistant MCF-7 cell was selected by poly-hema-coated plate. Growth rate and invasiveness of anoikis-resistant MCF-7 cells were analyzed and compared with corresponding adherent cells. The cell cycle distribution and apoptosis were detected by flow cytometry. Results Compared with doxorubicin ginsenoside Rg3 significantly inhibited the growth of anoikis-resistant breast cancer cell MCF-7(P 0.01). The cell cycle percentage of anoikis-resistant MCF-7 was also changed after treatment with ginsenoside Rg3. The cell percentage in G0/G1 was increased significantly(P 0.05). Conclusion Ginsenoside Rg3 can inhibit the growth of anoikis-resistant breast cancer cell MCF-7. The underlying mechanism may be related with the cycle arrest in anoikisresistant MCF-7.
