摘要
甘薯品种岩 8- 6 的叶片组织依次经硫酸铵分级沉淀、甲壳素亲和层析及 Sephadex G-75分离纯化得到一种凝集素 ( Ipomoea Batatas Lectin,简称 IBL) .纯化的 IBL电泳显示一条蛋白质着色带 .IBL的亚基相对分子质量为 4 70 0 0 .IBL对兔红细胞有凝集作用 ,凝血活性可被 D-果糖明显抑制 .IBL 对热不稳定 ,加热到 70℃时 ,活性完全丧失 .IBL对碱敏感 .IBL含有 1 6种氨基酸 ,富含 Asp和 Glu,中性糖含量为 5.0 7% .
The extraction solution of the variety of Ipomoea Batatas was graded by ammonium sulfate. It was affinity chromatographied by the chitin and filtered by molecular sieve. Author obtained the IBL (Ipomoea Batatas Lectin) which showed a single band on PAGE. And by SDS PAGE author measured subunit relative molecular mass to be about 47KD. IBL had agglutination on blood cells of rabbits, its agglutination activity was stopped by D fructose. IBL was heat unstable and lost all activity at 70℃. It was sensitive to alkali. IBL contained 5 07% neutral sugar and 16 kinds of amino acid and was rich in Asp and Glu.
