胚胎中胚层间充质细胞参与小鼠胚胎原始心管的形成

摘要 目的探讨小鼠胚胎早期原始心管发育。方法对胎龄7.5-9 d小鼠胚胎心脏进行石蜡连续切片,连续切片进行HE染色,并用抗转录因子GATA4和抗α-平滑肌肌动蛋白(α-SMA)抗体进行免疫组织化学染色。结果头端中胚层间充质细胞与小鼠胚胎原始心管及心包腔脏壁中胚层相延续;心肌早期分化标志蛋白转录因子GATA4和α-SMA在原始心管部位高表达。结论位于神经沟和原肠之间的胚胎中胚层间充质细胞参与原始心管和心包腔脏壁中胚层的形成;原始心管和心包腔脏壁中胚层细胞的迁移和分化机制不同。 Objective To explore the early development of the mouse embryonic primary heart tube.Methods Serial sections of embryonic day(ED) 7.5-9 mouse hearts were stained by HE staining,and immunohistochemistry staining with antibodies against transcriptional factor GATA4 and α-smooth muscle actin(α-SMA).Results Mesenchymal cells were seen extending to the primary heart tube and splanchnic epithelium of the pericardial cavity in the embryonic mesoderm of the head.The markers transcriptional factor GATA4 and α-SMA indicating early differentiation of cardiomyocyte showed strong expression in the primary heart tube.Conclusion The embryonic mesenchyme cells between the neural groove and foregut participate in the formation of both primary heart tube and splanchnic epithelium of the pericardial cavity,but the mechanisms of migration and differentiation are different.
