磺脲类受体在2型糖尿病大鼠菌状乳头味蕾的表达研究 被引量:1

摘要 目的探讨快效促泌剂瑞格列奈(repaglinide)额外通过头期机制促进餐时胰岛素分泌的可能性。方法取雄性2型糖尿病Wistar大鼠60只,采集舌菌状乳头和胰腺组织,采用免疫组织化学染色和Western blot方法判定大鼠味蕾有无磺脲类受体(sulfonylurea receptor,SUR)1和SUR2的表达及分布情况,并以同只大鼠胰腺作为阳性对照。结果发现2型糖尿病大鼠舌菌状乳头味蕾细胞存在单一SUR1表达。结论瑞格列奈快速促胰岛素分泌作用可能存在经味蕾介导的头期分泌途径。 Objective To test the possiblity that a fast-onset promoting agent repaglinide may initiate prandial insulin secretion through the mechanism of cephalic-phase insulin release. Methods Sixty male Wistar rats with type 2 diabetes mellitus were executed after general an- esthesia. The fungiform papillae and pancreas tissues were separately collected. The immuno- histochemical staining was used to detect the expression and distribution of sulfonylurea receptor 1 ( SUR1 ) or sulfonylurea receptor 2 (SUR2) in rat taste buds. The pancreas tissues from the same rat were used as positive control. Results SUR1 was uniquely expressed in type 2 diabe- tes mellitus rat taste buds of fungiform papillae. Conclusion The rapid insulin secretion-pro- moting effect of repaglinide may be exerted through the cephalic-phase secretion pathway medi- ated by taste release.
