超高效液相质谱联用法研究柴胡特征指纹图谱 被引量:5

摘要 目的:建立柴胡的化学成分特征指纹图谱,为柴胡的质量控制及品种鉴定提供依据。方法:采用UPLC-MS联用技术,通过质谱信息结合文献报道,分离鉴定柴胡中化合物成分,在此基础上建立柴胡药材的特征指纹图谱,分析比较特征指纹图谱峰的相对峰面积;并使用柴胡皂苷c、a、d的对照品计算柴胡药材中相应成分的含量。结果:根据质谱信息共分离鉴定了7个皂苷类化合物并作为柴胡药材的特征指纹图谱峰,各个柴胡样本其特征峰的相对峰面积有显著差异,柴胡皂苷c、a、d的含量也有明显差别。结论:不同产地柴胡药材中各个化学成分含量不呈相关性,说明差异很大;利用该方法可以作为柴胡药材整体质量评价的依据,并能为柴胡药材建立其"身份卡片",从而达到临床用药的安全、可控,为实现中药现代化提供理论基础。 OBJECTIVE To established the chemical constituents featureprints of Bupleurum, to provide evidence for the quality control and the identification of variety of Bupleurum. METHODS The constituents of Bupleurum were detected by liq uid chromatography-mass spectrometry (LC-MS). and identified through analyzing the information from mass spectrometry and the references. On the basis of above results, the featureprints of Bupleurum was established and the relative area of thefeatureprints were analyzed and compared. Saikosaponln a, c and cl were cnosen as the reference substances for the determination of constituents of Bupleurum. RESULTS Seven steroid saponins were obtained and identified from Mass Spectral informa tion,which were considered as the featureprints peaks of Bupleurum. There were significant differences in the featureprints peaks. It was obviously different from the content of Saikosaponin a, c and d. CONCLUSION There were significant differences on the contents of the chemical constituents of Bupleurum of different areas. The method could be used for the holistic quality evaluation of Bupleurum, and to establish the card for Bupleurum in the order to ensure the safety and control of clinical medi cation, and provide the theoretical basis for the modernization of Traditional Chinese Medicine.
