协同刺激分子B7-H1、B7-H4和叉状头/翅膀状螺旋转录因子阳性调节性T细胞在胃癌中的表达及其临床意义 被引量:5

Expression and clinical significance of costimulatory molecules B7-H1, B7-H4 and forkhead box P3+ regulatory T cells in gastric carcinoma

摘要 目的 探讨协同刺激分子B7-H1、B7-H4和转录因子叉状头/翅膀状螺旋转录因子(Foxp3)在胃癌中的表达及其临床意义.方法 应用免疫组织化学染色法检测B7-H1、B7-H4和Foxp3在100例胃癌组织及癌旁组织中的表达,分析B7-H1、B7-H4和Foxp3的表达与临床病理参数的关系.结果 B7-H1、B7-H4在胃癌组织中高表达,阳性表达率分别为65.0%和71.0%,其表达水平与浸润深度、淋巴结转移及TNM分期有关,差异有统计学意义(P<0.05).Foxp3+调节性T细胞(Treg)在胃癌组织中高表达,并与淋巴结转移有关,差异有统计学意义(P<0.05).B7-H1、B7-H4的表达水平和Foxp3+ Treg细胞数量呈正相关,相关系数分别为:r=0.437,P<0.05;r=0.456,P<0.05.B7-H1、B7-H4及Foxp3高表达患者的预后差于低表达者(P<0.05).结论 B7-H1、B7-H4和Foxp3+ Treg细胞在胃癌组织中高表达,并与胃癌患者淋巴结转移和预后有关. Objective To investigate the expression of B7-H1,B7-H4 and forkhead box P3 (Foxp3) in gastric carcinoma and its clinical significance.Methods The expression of B7-H1,B7-H4 and Foxp3 was detected by using IHC in 100 gastric carcinomas,and the relationship between the expression of B7-H1,B7-H4 and Foxp3 with clinicopathological features was statistically analyzed.Results Both B7-H1 and B7-H4 showed high expression in gastric carcinoma tissues (65.0% and 71.0%).The expression of B7-H1 and B7-H4 proteins was positively correlated to the depth of tumor invasion,lymph node metastasis and TNM stage.The positive expression rate of B7-H1 and B7-H4 in the patient with less than 5-year survival was obviously higher than that in the patients with more than 5-year survival (P < 0.05).High expression of Foxp3 + regulatory T cells (Treg) cells in gastric cancer tissues was correlated to lymph node metastasis (P < 0.05).Statistical analysis also showed that the expression of B7-H1 and B7-H4 protein was positively correlated to the number of Foxp3 + Treg cells (r =0.437,P < 0.05 ; r =0.456,P <0.05).Conclusion B7-H1,B7-H4 and Foxp3 + Treg cells were up-regulated in gastric cancer tissues,and may be correlated to gastric carcinoma metastasis and prognosis.
