摘要
目的 通过量子点(QDs)即新型半导体免疫荧光标记试剂结合组织芯片技术检测p53及基质金属蛋白酶-2(MMP-2)蛋白在人胃癌组织中的表达,并与免疫酶组织化学法比较,探讨免疫荧光组织化学试剂量子点技术的有效性和实用性.方法 采用半导体免疫荧光标记试剂-量子点结合组织芯片技术(QDs-IHC)观察人胃癌组织芯片中p53及MMP-2蛋白的表达,并且检测p53及MMP-2蛋白在胃癌组织中的共表达,并与常规免疫酶法的结果比较.结果 新型QDs-IHC和常规的免疫组织化学(IHC)法检测分析显示:p53(胃癌组p53蛋白两种方法的阳性检测率分别为72.86%和65.71%)及MMP-2蛋白(胃癌组:MMP-2蛋白两种方法检测的阳性率为87.14%和82.86%)表达在对照组和胃癌组之间比较,差异有统计学意义(P<0.05).p53及MMP-2蛋白表达之间呈显著正相关(r =0.533,P<0.01).QDs-IHC检测2种蛋白表达的阳性率均高于IHC所检测的结果,但差异无统计学意义(P>0.05).结论 新型QDs-IHC和常规IHC两种方法可检测到p53及MMP-2蛋白在胃癌组织中的表达,QDs-IHC技术检测其灵敏度比IHC高,图片显色背景比IHC干净.p53及MMP-2在胃癌患者的癌组织中共同表达.
Objective In this experiment we used quantum dots (QDs) by new type of semiconductor immunofluorescence labeling reagents combined with tissue chip technology to detect p53 and matrix metalloproteinase (MMP)-2 protein expression in gastric cancer tissue of the situation,and compared with the immune enzyme network stations,the immunofluorescence histochemical reagent validity and practicability of the quantum dots.Immunofluorescence histochemical.Methods Quantum dots immunohistochemistry (QDs-IHC) and p53 in human gastric cancer tissue microarray technology analysis and the expression of MMP-2 protein,and also tested the p53 and MMP-2 protein expression in the tissue of gastric cancer were,and compared with the conventional immune enzymatic results.Results New immunofluorescence labeling reagents semiconductor quantum dots (QDs-IHC) combined with tissue microarray technology and past conventional IHC testing analysis showed:p53 and MMP-2 protein expression in the control group,(The Positive expression rate of p53 are 72.86% and 65.71%,the Positive expression rate of MMP-2 are 87.14% and 82.86%),and compared between gastric cancer group,the differences were significantly (P <0.05).Between p53 and MMP-2 protein expression was significantly positive correlation (r=0.533,P<0.01).QDs-IHC testing two kinds of protein expression positive rate were higher than IHC testing results,but there was no significant difference (P > 0.05).Conclusion New immunofluorescence labeling reagents semiconductor quantum dots (QDs-IHC) combined with tissue microarray technology and conventional enzyme immunohistochemical (IHC) in the two methods can detect p53 and MMP-2 protein expression in gastric cancer tissue,through the QDs-IHC testing its sensitivity is higher than IHC technology,image color background is cleaner than IHC.The p53 and MMP-2 in the common expression in cancer tissue of patients with gastric cancer that different protein localization at the same time,the role to play in promoting malignant progression of gastric cancer.
