摘要
[目的]为科学保存和利用贵州李资源提供理论依据。【方法】采用ISSR分子标记技术对45份贵州李资源和3个美国引进品种进行遗传多样性分析,并进行亲缘关系鉴定。[结果]27条引物共扩增出198条DNA谱带,其中多态性条带181条,占扩增总带数的91.41%。供试材料Nei’s遗传多样性指数(H)为0.0412-0.4992,Shannon信息多样性指数(I)为0.1021-0.6931,遗传相似系数为0.50~0.93,表明贵州李资源遗传多样性丰富。在相似系数0.67处,48份李资源被聚为6组.其中2份鸡心李资源、2份九阡李资源、3份玉皇李资源以及4份酥李资源分别为同名异物。【结论】贵州李资源具有丰富的遗传多样性;采用该分子标记技术区分了同名异物的李资源。
[Objective]The aims of this study were to provide a scientific basis for the conservation and utilization of plum accessions in Guizhou. [Method]Genetic diversity and relationship of 45 plum accessions in Guizhou and 3 plum accessions introduced from the United States were evaluated by ISSR-PCR. [Result]The PCR by 27 ISSR primers yielded a total 198 bands and 181 seorable polymorphie markers,the number of polymorphie bands accounted for 91.41% of total bands.The Nei's genetic diversity index (H) was 0.041 2 to 0.499 2, the Shannon index of diversity (I) was 0.102 1 to 0.693 1, and the genetic similarity coefficient among all plum accessions ranged from 0.50 to 0.93, indicating that the plum accessions in Guizhou had abundant genetic diversity. 48 plum accessions could be classed into six groups at a Jaeeard coefficient of 0.67, and 2 varieties of Jixinli, 2 varieties of Jiuqianli, 3 varieties of Yuhuangli and 4 varieties of Suli were proved to be homonyms,respectively. [Conclusion]The substantial genetic diversity was identified among the plum accessions in Guizhou, and the plum accessions of homonym eould be differen- tiated by ISSR markers successfully.
