真武汤调节肾间质纤维化大鼠NF-κB表达的研究 被引量:12

Effect of Zhenwu decoction on nuelear factor kappa B expression in renal tubulointerstitial fibrosis in unilateral ureteralobstruction( UUO) rats

摘要 目的研究真武汤对NF-κB蛋白表达的影响,从分子水平对真武汤治疗肾间质纤维化的机制进行探索。方法采用单侧输尿管结扎的方法(UUO)制备大鼠肾间质纤维化模型,将150只SPF级健康雄性wistar大鼠,随机分为假手术组、模型组、尿毒清组以及真武汤低、中、高剂量组。术后2周、3周、4周分别处死大鼠,收集尿液、血清,结扎单侧肾组织,对24 h尿蛋白、血肌酐、尿素氮含量进行检测,分别用免疫组织化学法、ELISA法检测NF-κB的表达。结果在各时间点,真武汤各给药组与模型组相比,大鼠梗阻侧肾脏肿大和积水程度较轻,血肌酐、尿素氮含量较低,肾组织和血清中NF-κB蛋白表达均较弱。结论真武汤可减少间质纤维化,减轻肾组织损伤,其延缓肾间质纤维化进程的作用机制可能是通过抑制NF-κB的表达来实现的。 Objective To investigate the effect of Zhenwu decoction on nuelear factor kappa B expression of renal tubulointerstitial fibrosis in unilateral ureteralobstruction ( UUO) rats,and to explore the mechanism of the Zhen-wu decoction for treating renal interstitial fibrosis .Methods The renal interstitial fibrosis induced by unilateral ure-teral obstruction in rats was prepared .The 150 male wistar rats were randomly divided into six groups:sham operation group(n=21),UUO group (n=21),low-dosage of Zhenwu decoction group (n=27),medium-dosage of Z hen-wu decoction group (n=27),high-dosage of Zhenwu decoction group (n=27)and Niaoduqing granules group (n=27).The rats were given daily by gavage starting one day after operation .A part of rats in each group were sacrificed respectively at 2th、3th、4th week after operation .The 24h urine and serum were collected ,and the ligation of Unilater-al kidney tissues was conducted .The level of 24 hour urinary protein , serum creatinine ( Scr ) , blood urea nitrogen ( BUN) were detected ,and the expressions of NF -κB were observed by immunohistochemical method and ELISA . Results The levels of serum Scr and BUN in Zhenwu decoction groups were lower than those in UUO group at each indicated time points .The expression of NF-κB in blood and renal were much lower than that in the Zhenwu decoc-tion groups.Conclusion The Zhenwu decoction could delay the progression of renal interstitial fibrosis ,which may be involved in the decrease of NF -κB expressions in UUO rats .
