摘要
[目的]为肝素钠的大规模生产提供依据。[方法]利用3种复合酶从肠渣中提取肝素钠,通过测定提取肝素钠的质量、蛋白含量以及效价对3种复合酶法进行评价。[结果]由木瓜蛋白酶和链蛋白酶复合酶提取的肝素钠中蛋白含量最高(47%),效价为15USPU/mg;由胰蛋白酶和链蛋白酶复合酶提取的肝素钠中蛋白含量为29%,而其效价为34 USPU/mg。由2709碱性蛋白酶和胰蛋白酶复合酶提取的肝素钠中蛋白含量最少(24%),效价最高(39 USPU/mg)。[结论]2709碱性蛋白酶和胰蛋白酶复合酶能有效提取肠渣中残留的肝素钠。
[ Objective ] The research aimed to provide basis for large-scale production of heparin sodium. [ Method ] Heparin sodium was ex- tracted from intestinal contents by three kinds of compound enzymes. And the weight, protein content and titer of heparin sodium were deter- mined to evaluate three kinds of compound enzymes. [ Result] The protein content was the highest in heparin sodium extracted with compound enzyme of papain and pronase, and its titer was USPU/mg. The protein content in heparin sodium extracted with compound enzyme of trypsi- nase and pronase was 29% ,and its titer was 34 USPU/mg. The protein content was the lowest in heparin sodium extracted with compound en- zyme of 2709 alkaline protease and trypsinase was the lowest, and its titer was the highest(39 USPU/mg). [ Conclusion] The residual heparin sodium could be effectively extracted from intestinal contents bv comnound enzyme of 2709 alkaline protease and trypsinase.
