缺氧诱导因子-1α和血红素加氧酶-1在胃癌组织中的表达和临床意义 被引量:1

摘要 目的 研究缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)和血红素加氧酶-1(heme oxygenase-1,HO-1)在胃癌组织中的表达,探讨它们在胃癌发生发展、浸润和转移中的作用.方法 应用免疫组织化学SP法检测96例胃癌组织中HIF-1α和HO-1的表达,分析其临床意义及两者之间的相关性.结果 胃癌组织中HIF-1α和HO-1的阳性表达率为73.96%和70.83%,明显高于正常胃组织中的表达水平,差异有统计学意义(P〈0.05).HIF-1α和HO-1的表达与胃癌的不同组织学分级、临床TNM分期、浸润深度和淋巴结转移有关(P〈0.05或P〈0.01),与患者的年龄、性别和肿瘤的大小无关.HIF-1α和HO-1间存在相关性(r=0.657,P〈0.05).结论 HIF-1α和HO-1在胃癌发生发展中起重要作用,HIF-1α和HO-1的阳性表达可作为判断肿瘤具有高度侵袭转移潜能和预后不良的指标. Objective To detect the expressions of hypoxia-inducible factor-la (HIF-1α) and heine oxygenase-1 (HO-1) in gastric cancer and examine their correlations with occurrence, progres- sion, invasion and metastasis of gastric cancer. Methods Immunohistochemical technique was used to detect the expressions of HIF-1α and HO-1 in 96 cases with gastric cancer. The associations between the expressions of HIF-Ia and HO-I and clinicopathological features and prognosis were examined. Results The percentages of HIF-1α and HO-1 protein positive expression were 73. 96% and 70. 83% in gastric cancer respectively. And the expressions of HIF-1α and HO-1 were significantly higher in gastric cancer than those in normal gastric mucosa tissue(P〈0. 05). And their expressions in gastric cancer were significantly associated with TNM stage, invasive depth and lymph node metastasis(P〈0. (15 or P〈0. 01 )except for patient age, gender and tumor size. A positive correlation existed between the expressions of HIF-1α and HO-1 (r = 0. 657, P〈0. 05). Conclusions Both HIF-la and HO-I play important roles in the development of gastric cancer. And their expression profiles of HIF-1α and HO-I may serve as important prognostic indices of predicating tumor potentials in invasion and metas- tasis.
