摘要
目的研究通过与罗氏固体药敏法和BD960自动检测系统比较,判断三磷酸腺苷(ATP)发光法全新且检测快速简便试验方法的可行性。方法罗氏分枝杆菌药物敏感性测定按照《临床技术操作规范结核病分册》操作,BD960药敏试验操作按照《BD BACTEC MGIT 960system全自动分枝杆菌检测系统》培训手册操作,采用ATP发光法药敏试验操作,同时对147株临床菌株进行检测,并对结果进行比较。结果 ATP发光法检测结果与罗氏固体药敏法的符合率为95.2%,差异无统计学意义,与BD960自动检测系统的符合率为93.9%,差异无统计学意义;ATP发光法的检测时间为(6.6±2.1)d,比罗氏固体药敏法缩短很多,差异有统计学意义(t=418.0,P<0.01),略快于BD960自动检测系统(7.9±2.6d);ATP发光的只需要一台HygienaPI-102荧光检测仪,检测成本较低。结论 ATP发光药敏检测法快速简便、准确率高且成本低,其有效的避免了罗氏固体法过长的检测时间和BD960自动检测系统过高的检测成本,适合常规试验室开展。
OBJECTIVE To compare the adenosine triphosphate ( ATP) bioluminescence assay with Roche solid susceptibility method and BD960 automatic detection system to determine the feasibility of ATP bioluminescence assay in the rapid detection. METHODS The drug susceptibility was determined with Roche solid susceptibility method according to the Clinical Techniques and Operation Standards for Tuberculosis, the BD960 drug susceptibility testing was performed according to the training manuals of BD BACTEC MGIT 960 system automatic Mycobacterium detection system, and the drug susceptibility testing was carried out by means of ATP bioluminescence assay. Totally 14 7 strains of clinical isolates were detected at the same time, and the results were compared. RESULTS The accordance rate between the ATP bioluminescence assay and the Roche solid susceptibility method was 95. 2 %, without statistically significant diffeneace. The accordance rate between the ATP bioluminescence assay and the BD960 automatic detection system was 93. 9 %, without statistically significant diffeneace. The detection time of the ATP bioluminescence assay was ( 6. 6 ± 2. 1) days was significantly shorter than that of the Roche solid susceptibility method(t=418. 0,P〈0. 01), was slightly shorter than that of the BD960 automatic detection system(7. 9±2. 6) days. The ATP bioluminescence assay only needed a HygienaPI- 102 fluorescence detector with the lower cost. CONCLUSION The ATP bioluminescence assay has the advantages of rapid and simple operation, high accuracy and lower cost and effectively avoid the prolonged detection time as the Roche solid susceptibility method as well as the high cost of detection as the BD960 automatic detection system and it is suitable for the routine laboratory test.
