兔颈动脉外膜剥离血管出血的动物模型

摘要 目的建立一个稳定的血管外膜剥离出血的动物模型,用于临床进一步研究局部止血药物的效果。方法取健康新西兰大白兔12只,随机按胶原酶浓度分为1 g/L、2 g/L、4 g/L三组,胶原酶消化兔颈动脉至动脉外膜疏松,眼科镊钝性剥离外膜,在剥离过程中出现血管渗血,观察对比不同浓度胶原酶消化后动脉出血量的变化,并取消化前后的颈动脉行HE染色,光镜下观察外膜剥离程度,以确定胶原酶最佳消化浓度及剥离程度。结果 1g/L的胶原酶消化动脉外膜不彻底,无法造成动脉稳定的出血,4 g/L的胶原酶导致血管消化过度破裂出血,2 g/L的胶原酶消化血管外膜+眼科镊钝性剥离可使血管外膜剥离彻底,血管出血稳定。结论胶原酶消化+眼科镊钝性剥离血管外膜的方法可建立稳定的血管出血模型,其最佳消化浓度为2 g/L。 Objective To provide a stable model of bleeding for further study on the clinical effect of topical hemostatic drugs, and try to establish a stable animal model of vascular tunica adventitia stripping of bleeding. Methods A total of 12 healthy New Zealand rabbits, according to the concentration of collagenase were randomly divided into three groups, 1 g/L, 2 g/L, 4 g/L. Different concentrations of collagenase digest rabbit carotid artery to artery tunica adventitia loose. Tuniea adventitia was blunt dissected by ophthalmic forceps. In the process of stripping oozed blood vessels, the changes of the bleeding after comparing different collagenase concentration digestion of tunica adventitia were observed. Then the rabbit carotid artery was taken into HE staining and the tunica adventitia by light microscope was observed to determine the best collagenase digestion concentration and the degree of tripping. Results Tunica adventitia was not com- pletely digested in 1 g/L collagenase concentration. It could not cause stable hemorrhage. But it could be excessive di- gested in 4 g/L collagenase concentration and cause uncontrolled bleeding. Tunica adventitia was completely digested in 2 g/L collagenase concentration and could cause stable bleeding by blunt dissection. Conclusion Collagenase diges- tion and blunt dissection of tunica adventitia can establish stable vascular hemorrhage model, the best concentration of digestion is 2 g/L.
