摘要 目的通过外用2,4-二硝基氟苯(DNFB)丙酮/橄榄油溶液致敏与激发BALB/c小鼠,探索特应性皮炎(atopic dermatitis,AD)模型的建立方法。方法将16只小鼠随机分为模型组及对照组各8只。实验第1天和第2天在模型组和对照组小鼠背部脱毛区分别涂搽一次DNFB丙酮/橄榄油溶液及丙酮/橄榄油,第7天开始每隔2天涂搽一次,共涂搽10次。观察小鼠皮肤的生理和病理改变。结果外用DNFB致敏与激发小鼠完成后,小鼠搔抓频率增加,小鼠皮肤可见慢性皮损改变,镜下可见慢性炎细胞浸润,ELISA血清检查发现IgE、IL-4及IFN-γ在血清中高表达。结论外用DNFB致敏与激发BALB/c小鼠产生的皮肤生理和病理改变与AD患者一致,可作为研究AD的发病机理及探索其治疗手段的有效的动物模型。 Objective To establish a mouse model of atopic dermatitis (AD)by sensitizing and stimulating BALB/c mice with 2,4-Dinitrofluorobenzene(DNFB) in acetone/olive oil. Methods Sixteen BALB/c mice were randomly divided into model and control groups ,8 in each group. On the first and second day of experiments, DNFB in acetone/olive oil were applied to an animal back area with hair removal in the model group. Then,the application was performed every two days on the seventh day of experiment,and 10 times in total. Meanwhile, mice in the control group were only dealt with acetone/olive oil. The changes of mice skin were observed. Results In the model group, scratching frequency of mice was increased. In these animals chronic skin lesions were observed, and a chronic inflammatory cell infiltration was found under microscopy. High expressions of IL-4, IFN-γ and lgE in serum were also detected. Conclusion The changes in the animal model were very similar to those in human AD. Therefore, the mice model can be used for elucidating the pathogenesis and exploring the effective treatment of the disease.
