摘要
目的了解microRNA-21(miR-21)在急性B淋巴细胞白血病(B-ALL)骨髓单个核细胞(BMMCs)中的表达水平及其意义。方法收集36例B-ALL病儿(其中高危型13例,标危型23例)化疗前后及12例骨髓常规正常非造血系统疾病病儿(对照组)的骨髓标本,采用实时荧光定量PCR方法检测BMMCs中miR-21表达。结果化疗前B-ALL组miR-21表达水平明显高于对照组,高危型组miR-21表达水平明显高于标危型组,差异有统计学意义(t=20.75、23.23,P<0.05)。化疗后B-ALL病儿miR-21表达水平较化疗前明显减低(t′=10.56,P<0.05),其中以化疗敏感组减低最为明显(t=13.57,P<0.05);化疗耐药组miR-21表达水平化疗前后无明显变化(P>0.05)。结论 miR-21过表达在B-ALL发病中发挥着重要作用,可作为判断儿童B-ALL预后指标之一。
Objective To assess the expression of microRNA-16(miR-21)and its significance in bone marrow mononuclear cells(BMMCs)in children with acute B-lymphocytic leukemia(BLL). Methods Bone marrow samples of 36kids with acute BLL(13with high risk and 23with standard risk,before and after chemotherapy)were collected,and 12kids of normal bone marrow picture with disesses of non-hematopoietic system served as controls.The expressions of miR-21in BMMCs were detected by real-time polymerase chain reaction. Results Before chemotherapy,the expression of miR-21in the acute BLL group was higher that that in the control,that of high risk type was higher than the standard risk tipe(t=20.75,23.23;P0.05);After chemotherapy,the expression level of miR-21in the BLL was lowered as compared with before,especially obvious in that with chemotherapy-sensitive(t=13.57,P0.05).For those with chemotherapy-resistant,the expressions of miR-21before and after therapy were not significantly different(P0.05). Conclusion The over-expression of miR-21plays an important role in the development of childhood acute BLL,which can be used as one of the markers to predict prognosis of the condition.
