一株云杉八齿小蠹伴生蓝变真菌的鉴定及致病性测定 被引量:1

摘要 [目的]确定1株云杉八齿小蠹伴生蓝变真菌C59的分类地位。[方法]对分离自伊春市五营区黑龙江丰林国家级自然保护区红皮云杉林内的一株云杉八齿小蠹伴生蓝变菌株进行培养特性、形态学特征观察和18S rDNA ITS序列分析鉴定,在对该蓝变菌株进行接种的同时,测定了该菌株的致病性,苗木感病后韧皮部的水分代谢,针叶叶绿素含量,韧皮部可溶性糖、蛋白质、MDA和脯氨酸含量。[结果]该蓝变菌株为Ophiostoma sp.,该菌株使接种苗木韧皮部发生蓝变,影响苗木韧皮部的水分代谢,导致苗木针叶叶绿素含量、韧皮部可溶性糖含量、韧皮部蛋白质含量下降,使苗木韧皮部MDA含量和脯氨酸含量上升。[结论]分离自伊春市五营区黑龙江丰林国家级自然保护区红皮云杉上的云杉八齿小蠹伴生蓝变菌株为Ophiostoma sp.,该菌株对红皮云杉苗木具有很强的致病性。 [ Objective] The taxonomic status of blue-stain fungal strain associated with Ips typographus isolated from Picea koraiensis was de- termined. [ Method ] The blue-stain fungal strain associated with Ips typographus isolated from Picea koraiensis in Heilongjiang Fenglin Nation- al Nature Reserve of Wuying district of Yichun city, was identified as Ophiostoma sp. based on culture characteristics, morphological features and 18S ITS sequence analysis. [ Result] The result of pathogenicity test showed that the blue-stain fungal strain can make the phloem color of the inoculated seedlings' turn to blue; affect water metabolism of seedling phloem; cause chlorophyll content, soluble sugar content and pro- tein content decreased ; and led MDA levels and proline content increased. In conclusion, Ophiostoma sp. [ Conclusion ] The blue-stain fungal strain associated with Ips typographus, isolated from Picea koraiensis in Heilongjiang Fenglin National Nature Reserve of Wuying district of Yi- chun city has a strong pathogenicity to Picea koraiensis.
