双曲霉混合发酵豆渣产纤溶酶的研究

Studies on the Fibrinolytic Enzyme Produced by Two Mixed Aspergillus Fermenting Soybean Residue

摘要 以豆渣为发酵基质,对甲基苯磺酰-L-精氨酸甲酯(TAME)底物法为检测方法,纤溶酶活性为检测指标,研究米曲霉3.800和黑曲霉3.430 9双曲霉混合发酵产纤溶酶的最佳工艺条件。通过单因素试验确定:米曲霉3.800和黑曲霉3.430 9双曲霉混合发酵最适接种量为10%,最适黑曲霉3.430 9与米曲霉3.800比例为1︰4,最适发酵时间为72h,最适碳源为麸皮。利用Box-Behnken设计优化培养基后,当碳氮质量比(C/N)为0.25,pH 7.2,含水量为76%时,发酵液中纤溶酶实际活性为1.265 3 TAME/mL。 Soybean residue as cultural medium, Na-4-tosyl-L-arginine methyl ester hydrochloride(TAME) as the substmte of fibrinolytic enzyme, fibrinolytic enzyme activity as detection index, the best fermentation conditions on the fibrinolytic enzyme produced by two mixed Aspevgillus fermenting soybean residue were studied. The single-factor experiments result showed the optimal conditions for fermentation were: 10% of inoculum, the proportion ofAsperillus oryzae and Aspergillus niger is 1 : 4, for 72 h, bran as carbon source. With the Box-Behnken design to optimize medium, the fibrinolytic enzyme activity reached 1.265 3 TAME/mL. Under the optimal conditions, the proportion of bran and soybean residue (C/N) was 0.25, the beginning pH and moisture content respectively was 7.22 and 76%.
