摘要
目的:研究痫宁喷雾剂对氯化锂-匹罗卡品致痫大鼠海马神经元损伤及c-fos表达的影响,探讨其治疗癫痫的机制。方法:将46只SD大鼠随机分为空白组、模型组、对照组和鼻腔组。采用氯化锂-匹罗卡品制成慢性癫痫大鼠模型,分别给予生理盐水喷鼻,丙戊酸钠灌胃,痫宁喷雾剂鼻腔给药。病理切片观察大鼠海马神经元损伤情况,免疫组化法检测大鼠海马c-fos的表达。结果:对照组与鼻腔组大鼠海马神经元水肿、固缩等损伤较模型组明显减轻(P<0.01),对照组与鼻腔组大鼠海马c-fos表达较模型组明显下降(P<0.05),鼻腔组与对照组比较无显著性差异(P>0.05)。结论:痫宁喷雾剂能明显减轻癫痫大鼠海马神经元水肿、固缩等损伤,降低c-fos表达水平,对神经元具有保护作用,其作用机制可能与降低神经元c-fos的表达有关。
Objective: To study the influence of xianning nasal spray on the injury of neuron and the expres- sion of c-los in the hippocampus of epileptic rats induced by lithium-pilocarpine, and explore its anti-epileptic mechanism. Methods: 46 SD rats were randomly divided into blank group, model group, contrast group and nasal cavity group. The chronic epileptic rat model was induced by lithium-pilocarpine. These groups were respectively given normal saline by nasal spray, VPA by gavage and Xianning nasal spay by intranasal administration. The neuron damage was observed by no^nal pathological means, and the expression of c-los in rats'brain was detected by immunohistochemical techniques. Results : The neuron damage of contrast group and nasal cavity group was ob- viously lightened compared with model group ( P 〈 0.01 ). The expressions of c-fos of contrast group and nasal cavity group were remarkably reduced compared with model group ( P 〈 0.05 ). The difference between nasal cavi- ty group and contrast group has no statistical significance (P 〉 0.05 ). Conclusion: Xianning nasal spray can a- bate the neuron damage in epileptic rats induced by lithium-pilocarpine, and downregulate the expression of c-fos in hippocampus, and protect the brain. Its antiepileptic mechanism might be correlated to the inhibition of c-los expression.
