高效液相色谱法测定不同产地棘托竹荪中4个核苷含量 被引量:7

摘要 目的:运用高效液相色谱法分析棘托竹荪中尿苷、肌苷、鸟苷和腺苷的含量。方法:采用GracePrevailSeleet C18(4.6mm×150mm,3μm)分析柱,以水(A)-甲醇(B)为流动相二元梯度洗脱(0~6.0min。0→5%B;6.0-15.0min,5%B→25%B;15.0—20.0min,25%B→100%B),流速1mL·min^-1,检测波长254nm,柱温25℃,进样量10乩。结果:尿苷、肌苷、鸟苷和腺苷浓度分别在2.0—181.0μg·mL^-1(r=1.000)、1.5—190.0μg·mL^-1(r=0.9999)、0.8-126.4μg·mL^-1(r=0.9999)和1.0—171.2μg·mL^-1(r=1.000)范围内线性关系良好,加样回收率(n=6)分别为100.7%(RSD=3.4%)、98.5%(RSD=4.2%)、99.5%(RSD=3.6%)和99.1%(RSD=3.8%);采自不同产地的棘托竹荪核苷含量差别较大,四川青川木鱼产的竹荪中尿苷、肌苷、鸟苷和腺苷4个核苷类成分含量是所有测试样品中最高的,含量分别为0.77、0.06、1.19和1.60mg·g^-1。结论:本法能够快速、简便地测定棘托竹荪中主要核苷的含量.为其质量控制提供保证. Objective : To determine nucleosides (uridine, inosine, guanosine and adenosine) in Dictyophora echi- novolvata by HPLC - DAD. Methods: The Grace Prevail Select Clscolumn (4. 6 mm x 150 mm,3 μm) was used for simultaneous determination of uridine, inosine, guanosine and adenosine in D. echinovolvata. Gradient elution with water (A) and methanol ( B ) was performed as follows :0 - 6.0 min,0→5 % B ;6. 0 - 15.0 min, 5 % B→25 % B;15.0 -20.0 min,25%B→100%B. The flow rate was 1 mL · min-1 and the injection volume was 10 μL. The system operated at 25 ℃ and the 4 nucleosides were monitored and quantified at a wavelength of 254 nm. Results: Uridine,inosine, guanosine and adenosine showed good linearity in the ranges of 2. 0- 181.0 μg ·mL^-1 (r = 1. 000 ), 1.5 - 190. 0μg ·mL^-1 ( r = 0. 9999 ) ,0. 8 - 126. 4μg ·mL^-1( r = 0. 9999 ) and 1.0 - 171.2 μg ·mL^-1 (r = 1. 000) ,and the average recoveries (n =6) were 100.7% (RSD =3.4% ) ,98.5% (RSD =4.2% ) ,99.5% (RSD = 3.6% ) and 99. 1% (RSD = 3.8% ) ,respectively. The contents of investigated nucleosides in D. echino- volvata from different production areas were different. The contents of the four nucleosides (uridine, inosine, guanosine and adenosine) in D. echinovolvata from Qingehuan Muyu of Sichuan Province were the highest among the 5 samples, their eontents were 0.77,0.06,1.19 and 1.60 mg · g^-1, respectively. Conclusion: The HPLC method established in this study is a rapid and simple method for the simultaneous determination of nueleosides in D. echinovolvata and is helpful to its quality control.
