HPLC法测定柘木中槲皮素、染料木素和山柰素的研究 被引量:6

摘要 目的:采用高效液相色谱法同时测定柘木中3个黄酮类成分(槲皮素、染料木素和山柰素)的含量。方法:采用ApolloC18色谱柱(4.6mm×250mm,5μm),以1%醋酸一甲醇为流动相,梯度洗脱(0~28min,65%A→40%A);流速:1mL·min^-1;柱温:30℃;检测波长:260nm。结果:槲皮素、染料木素和山柰素进样量分别在0.159~1.59wg(r=0.9997)、0.046—0.46μg(r=0.9995)和0.048~0.48μg(r=0.9998)范围内线性关系良好;平均回收率(n=6)分别为98.2%(RSD=2.6%)、100.6%(RSD=2.9%)和100.4%(RSD=2.7%)。结论:本法为该药材质量控制研究提供了参考。 Objective: To develop a method for determining 3 flavonoid constituents (quercetin, genistein and kaempferol) in Cudrauia tricuspidata. Methods:The analysis was conducted on an Apollo C18column (4. 6 mm × 250 mm, 5 μm) eluted with 1% acetic acid and methanol as mobile phases in gradient mode (0 -28 min ,65 % A→40% A). The flow rate was 1 mL · min-1 and the detection wavelength was set at 260 nm. The column temperature was 30 ℃. Results:The linear concentration ranges of quercetin, genistein and kaempferol were 0. 159 - 1.59 p,g (r =0. 9997) ,0. 046 -0. 46 μg (r =0. 9995) and 0. 048 -0. 48 p,g (r =0. 9998) ,respectively;The average re- coveries ( n = 6) were 98.2% ( RSD = 2. 6% ), 100. 6% ( RSD = 2.9% ) and 100. 4% ( RSD = 2.7% ), respec- tively. Conclusion:The method is simple, accurate and sensitive, and can be used for the quality control of Cudra- nia tricuspidata as a reference.
