养阴透解法对SLE小鼠脾脏组织IL-4和IFN-γ基因表达的影响

Effect of Yangyin Toujie therapy on the gene expression of IL-4 and IFN-γ in spleen tissue of mice with SLE

目的 探讨系统性红斑狼疮小鼠的免疫失衡状态以及清养透解法调节该平衡的效果.方法 将24只8 ～ 10周龄雌性MRL/lpr狼疮小鼠按质量随机分为A组、B组、C组、D组,分别给予清养透解合剂、泼尼松、清养透解合剂＋泼尼松、生理盐水灌胃进行干预,每组6只;另取8 ～10周龄雌性BALB/C小鼠6只作为E组,给予生理盐水灌胃.6周后眼眶取血检测T淋巴细胞亚群CD3、CD4、CD8的比例及调节性T细胞CD4＋ CD25＋的数量;取脾脏组织并采用RT-PCR法检测小鼠脾脏组织IFN-γ及IL-4基因表达情况.结果 ①与E组比较,D组IFN-γ水平明显下降（P＜0.01）,A、B、C组IFN-γ水平亦有显著下降（P均＜0.05）;A组、B组、C组两两比较无显著性差异（P＞0.05）.②与E组比较,D组IL-4水平明显上升（P＜0.01）,A组、B组、C组与D组比较均有显著性差异（P均＜0.05）,A组、B组、C组、E组两两比较无显著性差异（P均＞0.05）.③与E组比较,A组、D组Th1/Th2比值明显降低（P均＜0.01）,C组与E组比较无显著性差异（P＞0.05）,B组与E组比较有显著性差异（P＜0.05）;A、B、C组与D组比较均有显著性差异（P均＜0.05）.结论 MRL/lpr小鼠存在Th1/Th2细胞平衡紊乱,运用清养透解合剂配合西药的中西医结合疗法治疗SLE效果明显优于其他疗法.

Objective It is to explore the immune imbalance in mice with SLE and the effect of Yangyin Toujie therapy on this imbalance.Methods 24 female MRL/lpr mice with 8-10 weeks old were randomly divided into group A,B,C,D with 6 mice in each group according to body weight and respectively given Yangyin Toujie mixture,prednisone,angyin Toujie mixture ＋ prednisone,normal saline by intragastric administration.6 female BALB mice were chosen additionally as group E and given normal saline by intragastric administration.After 6 weeks blood was gotten from orbital cavity to determine T-lymphocyte subsets CD3,CD4 and CD8 and regulatory T cell CD4,CD25.The expression of IFN-γand IL-4 in spleen tissue was detected by RT-PCR.Results ①Compared with group E,the level of IFN-γ obviously decreased in group D （P ＜ 0.01）,the levels also significantly decreased in group A,B,C （P ＜ 0.05） ; compared with each other,there was no significant difference among group A,B,C（P ＞0.05）.②Compared with group E,the level of IL-4 obviously increased in group D（P ＜0.01）,compared with group D,the differences in the level were significant in group A,B,C （P ＜ 0.05）,there was no difference among group A,B,C,E（P ＞0.05）.③Compared with group E,the differences in Th1/Th2 in group A,D were much significant （P ＜ 0.01）,the difference in the ratio was not significant between group C and E （P ＞ 0.05） and significant between group B and E （P ＜ 0.05）,compared with group D,there were also differences in group A,B,C （P ＜ 0.05）.Conclusion MRL/lpr mice had Th1/Th2 cell imbalance,and Yangyin Qingyang therapy combined with western medicine had a better curative effect on SLE than the other treatment.
