西兰花种子中萝卜硫素粗提物稳定性研究 被引量:2

摘要 萝卜硫素是具有较强抗癌活性的物质,因此具有很高的研究价值。本实验研究了溶剂、温度、pH值、光照、抗坏血酸、金属离子对西兰花种子中萝卜硫素粗提物稳定性的影响。萝卜硫素粗提物在纯水、乙醇、丙酮、甲醇等4种不同溶剂、-4,4,20,40,60,80℃6种不同温度、pH 3,5,6.6,7,9,11等6种不同pH值、光照和黑暗、加抗坏血酸、不同金属离子即K+,Na+,Ca2+,Mg2+,Al3+,Fe3+处理下,分别于第0,2,4,6,8,10,20,30天取样,采用HPLC测定其萝卜硫素含量。pH对西兰花种子中萝卜硫素粗提物稳定性影响最大,碱性条件促进其降解,因此保存在酸性偏中性的条件下;温度对萝卜硫素粗提物稳定性影响次之,保存时应在低温-4℃以下条件下;而其他光照条件、抗坏血酸添加、金属离子对其稳定性影响不大。西兰花种子提取萝卜硫素工艺过程及提取物贮存条件建议为:溶剂为甲醇或丙酮、温度在-4℃以下低温、酸性偏中性、黑暗下贮存。 Sulforaphane is the material with strong anti-cancer activity,so it has high research value.We studied the stability of sulforaphane crude extract with solvent,temperature,pH,illumination,ascorbic acid and metal ions.The sulforaphane crude extract was treated with different solvents (pure water and ethanol and acetone and methanol),different temperatures (-4,4,20,40,60,80℃),different pH values (3,5,6.6,7,9,11),light and dark,adding ascorbic acid and different metal ions (K+,Na+,Ca2+,Mg2+,Al3+,Fe3+),then,the sulforaphane contents were determined by HPLC in the 0th,2nd,4th,6th,8th,10th,20th and 30th days.The pH value was the biggest influence factor of sulforaphane crude extract stability.Alkaline condition could promote sulforaphane to degrade,so it had better be saved in the acidic partial neutral condition.Temperature was the second influence factor of sulforaphane stability.So it should be preserved at or below-4℃.The other factors had few effects on the stability.The optimal condition for sulforaphane production process and storage from broccoli seed is that solvent:methanol or acetone,at or below-4℃,acid partial neutral environment and dark.
