摘要
目的以苯甲酸为对照品,测定白芍药材中白芍总苷的含量。方法采用碱水解-HPLC法测定白芍药材中苯甲酸的含量,以公式“白芍总苷含量%=苯甲酸含量%x480.27/122.12”计算白芍总苷的含量;采用Kromasil 100-5C18 250*46mm色谱柱,流动相为乙腈-0.1%磷酸(26:74),检测波长230nm,流速1mL·min-1,柱温30℃。结果方法重现性和稳定性均良好,苯甲酸在4.34—43.4ug·mL-1线性关系良好,回归方程为Y=59136.6879X+4034.7289,r=1.0000(n=5),平均回收率为97.88%,RSD为2.13%(n=6)。结论该方法用于白芍药材中白芍总苷的含量测定稳定、可靠。
Objective To set up a simple and accurate method for the determination of total glucosides of paeony in Radix Paeoniae Alba(TGP) with benzoic acid as control sample. Methods Using the method of alkaline hydrolysis-HPLC, the amounts of TGP were obtained from the following formula: TGP(%)=benzoic acid(%) ×480.27/122.12. Chromatographic conditions concluded 18 alkyl silane Bonded Silica as the stationary phase, a mixture of acetonitrile - 0.1% phosphoric acid (25:75) was the mobile phase, the detection wavength was 230nm, the flow rate was 1.0 mL/min, the column temperature was 30℃. Results The calibration curves of benzoic acid were in good linearity over the range of 4.34-43.4ug/mL(r=1.000 0). The average recovery rate was 97.88%, RSD was 2.13%(n=6). Conclusion The method was repeatable and stable, and it can be used to determine the total glucosides ofpaeony in Radix Paeoniae Alba.
