摘要
目的研究蛴螬生物碱的提取工艺并对其抗肿瘤活性进行评价。方法采用单因素实验和L9(34)正交实验得到最佳提取蛴螬生物碱的方法;采用MTT和荷瘤小鼠抑瘤率的方法测定生物碱的体内外抗肿瘤活性。结果最佳提取工艺为蛴螬粗粉用体积分数95%乙醇浸泡提取24h;蛴螬生物碱对体内外肿瘤细胞有明显抑制作用,且具有质量浓度依赖性。结论蛴螬粗粉用体积分数95%乙醇浸泡24h提取,浓缩浸膏经过酸化萃取得到的生物碱抗肿瘤活性显著。
Objective To optimize the extracting technique for alkaloids from Holotrichia diomphalia Bates and to evaluate their an-titumor activity .Method Single factor analysis and L9 (34 ) orthogonal test were used to optimize the extraction method of Ho-lotrichia diomphalia Bates alkaloid .The anti-tumor activities of alkaloids in vitro were evaluated in human cancer cells by MTT assay .S180 sarcoma mice were used for the anti-tumor test in vivo .Results The optimal condition was to dip the coarse powder of Holotrichia diomphalia Bates for 24 h in 95% alcohol .The alkaloids from Holotrichia diomphalia Bates inhibited human cancer cells and development of transplanted tumor in mice in a dose dependent manner .Conclusion The optimal condition was to dip the coarse powder of Holotrichia diomphalia Bates for 24 h in 95% alcohol .After filtrating condensing ,and acidification ,the extraction was performed .The alkaloid from Holotrichia diomphalia Bates showed significant anti-tumor activities .
