摘要
多烯大环内酯类抗生素是最有效的抗真菌素之一,在临床和食品工业中都得到广泛的应用。本文综述了多年来多烯大环内酯类抗生素的研究成果及进展。包括:多烯大环内酯类抗生素完整的生物合成流程;该类抗生素杀灭真菌核心机制的最新发现;通过生物工程手段对多烯大环内酯类抗生素进行结构改造的研究,目前人们已实现通过有目的性的理性设计,调整生物合成相关基因来获得预期产物;多烯大环内酯类抗生素从属的聚酮类抗生素相关基因的进化方式和特点等。在介绍多烯大环内酯类抗生素各个方面的研究进展的同时,还对多烯大环内酯类抗生素的研究与应用前景进行了展望。
Polyene macrolide antibiotics are one of the most effective antifungal agents, which are widely used in clinical and food industries. Through years of research, the complete biosynthetic processes of polyene macrolide antibiotics are fully explored. With the development of research, the core fungicidal mechanism is established and the traditional mechanism is proved to be a complementary mechanism. For study of polyene macrolide antibiotics engineering via bioengineering methods, foreign scholars managed to obtain desired product through rational design of biosynthetic genes. Studies on evolution of polyketide antibiotic containing polyene macrolide antibiotics reveal evolutionary characteristics of polyketide antibiotic. Based on many studies on polyene macrolide antibiotics as well as relevant research of our research group, the authors make a developmental prospect of polyene macrolide antibiotics.
