摘要
E0基因编码的蛋白是牛病毒性腹泻—黏膜病病毒(BVDV)的主要保护性抗原之一,其是研制BVDV基因工程疫苗的候选者。E0基因在卡介苗中难以表达,已有研究结果表明E0蛋白具有RNase活性。根据卡介苗密码子使用频率分析E0基因,发现其有多个稀有密码子。通过对该基因截短和密码子优化,在卡介苗中表达E0基因,检测到了E0蛋白的抗原活性,为构建可同时预防结核和牛病毒性腹泻—黏膜病的二价基因工程疫苗奠定基础。
The protein encoded by E0 gene is one of the major protective antigens of bovine viral diarrhea-mucosal disease vi- rus (BVDV). Eo gene is a candidate for developing genetic engineering vaccine of BVDV and it could not express in BCG, it has been shown that Eo protein has RNase activity, and has a plurality of rare codons according to analyze the codon usage frequen- cy of BCG. By truncating the gene and optimizing its codons, E0 gene expressed in BCG successfully and its antigenic activity was detected, which laid the foundation for building bivalent genetic engineering vaccine to prevent tuberculosis and bovine viral diarrhea-mueosal disease at the same time.
