摘要
为探讨泌乳乳腺中必需氨基酸与L型氨基酸转运载体1(L-type amino acid transporter 1,LAT1)表达之间的关系,本研究通过体外构建奶牛乳腺泌乳模型,采用免疫印迹方法检测生理浓度的必需氨基酸对乳腺腺泡中LAT1及其辅因子4F2重链(4F2heavy chain,4F2hc)表达变化的影响。结果表明,生理浓度的必需氨基酸显著上调体外培养的奶牛乳腺腺泡中LAT1和4F2hc的表达,提示LAT1是奶牛乳腺上皮细胞内负责转运必需氨基酸的主要转运载体,LAT1和4F2hc的表达受必需氨基酸的诱导。
To investigate the effect of essential amino acids on LAT1 expression in lactation mammary gland, the lactation mammary acini were cultured and LAT1 and 4F2hc expression were detected by Western blotting. The results showed that es- sential amino acids upregulated LAT1 and 4FZhc expression significantly in lactation mammary acini of dairy cow. It revealed that LAT1 was the mainly amino acid transporter in lactation mammary gland. The expression of LAT1 and 4F2hc was induced by essential amino acids.
