清脂通脉颗粒改善家兔血脂及抗动脉粥样硬化随机平行对照研究 被引量:4

摘要 [目的]探讨清脂通脉颗粒对家兔血脂、主动脉粥样硬化影响及机制。[方法]建立高脂家兔模型。将48只家兔按随机数字表法分为6组:空白对照组、模型组、血脂康组、清脂通脉颗粒低剂量组、清脂通脉颗粒中剂量组、清脂通脉颗粒高剂量组,8只/组。造模成功后,除空白对照组外,其它各组灌胃干预,15mL/kg·次,2次/d。模型组予生理盐水;血脂康组予血脂康0.1g/kg·d;清脂通脉颗粒各剂量组,分别为0.69g/kg·d(3g生药/kg·d),2.31g/kg·d(10g生药/kg·d)以及6.94g/kg·d(30g生药/kg·d)。连续灌胃干预10周。观测各组大鼠血清甘油三脂(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)。[结果]模型组4项血清值检测结果均高于空白对照组(P<0.05)。清脂通脉颗粒中、高剂量组甘油三酯(TG)均明显低于模型组(P<0.05),与实验用剂量的血脂康作用相近(P>0.05)。清脂通脉颗粒对总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)以及高密度脂蛋白胆固醇(HDL-C)未见明显影响(P>0.05)。主动脉光镜下病理学观察,空白对照组:动脉壁结构清晰,内膜光滑,内皮细胞完整连续,内弹性膜及中膜弹力纤维形态正常,未见病理变化,平滑肌细胞排列规则。模型组:内膜明显增厚,内膜表面不光滑,弹力纤维断裂,平滑肌细胞排列紊乱,大量泡沫细胞形成,可见大块脂质侵润斑块向管腔凸出,大量增生的炎性细胞。血脂康组:动脉壁结构较清晰,内膜较光滑,有少量的内皮细胞泡沫样改变,平滑肌细胞排列较规则,增生的炎性细胞减少。清脂通脉颗粒低剂量组:大部分结构清晰,内膜较光滑,偶见主动脉内皮细胞泡沫样改变,平滑肌细胞排列较规则,增生的炎性细胞减少。清脂通脉颗粒中剂量组:主动内膜较光滑,偶见少量轻微的内皮细胞泡沫样改变,平滑肌细胞排列较规则,炎性细胞减少。清脂通脉颗粒高剂量组:主动脉结构清晰,内膜较光滑,内皮细胞泡沫样改变消失,平滑肌细胞排列规则,炎性细胞明显减少。[结论]清脂通脉颗粒能降低血脂,抗动脉粥样硬化机制可能与降脂有关。 [ Objective ] Explore Qingzhi Tongmai lipid particles in rabbit aortic atherosclerosis and its mechanisms. [ Methods ] Fat rabbit model established model. The 48 rabbits were randomly divided into 6 groups randomly : control group , model group, Xuezhikang group, Qingzhi Tongmai particles low dose group, Qingzhi Tongmai particles dose group , QingZhi Tongmai particles high-dose groups of eight . After the modeling , in addition to the blank control group, the other groups according to the following dose gavage , 15mL/kg / time, 2 times / d, the end of the experimental period to 10 weekends . Model group, normal saline ; Xuezhikang I Xuezhikang 0.1g/kg ~ d; Qingzhi Tongmai particles low-dose group , middle dose group and high dose group were I Qingzhi Tongmai for the sample of particles at doses of 0.69g/kg · d ( 3g crude drug / kg / d ), 2.31g/kg · d ( 10g crude drug / kg / d ) and 6.94g/kg· d ( 30g crude drug / kg / d ) . Treatment for 10 weeks. After treatment, the serum triglyceride determination ( TG ), total cholesterol ( TC ), high density lipoprotein cholesterol ( HDL-C ), low density lipoprotein cholesterol ( LDL-C ) . [ Result ] Serum values of model group 4 test results were higher than the control group ( P〈0.05 ), the model was established that high blood lipids. Qingzhi Tongmai particles, the high-dose group triglyceride ( TG ) were significantly lower than the model group ( P〈0.05 ), described Qingzhi Tongmai particles can reduce high blood cholesterol rabbit serum triglycerides, and experiment with Xuezhikang dose effect similar ( P〉O.05 ) . Qingzhi Tongmai particles on total cholesterol ( TC ), low density lipoprotein cholesterol ( LDL-C ) and high-density lipoprotein cholesterol ( HDL-C ) had no significant effect ( P〉O.05 ) . Aortic pathology observed under light microscope, blank control group : the arterial wall structure is clear , intimal smooth, continuous endothelial cell integrity , internal elastic membrane and the membrane elastic fibers were normal , no pathological changes in smooth muscle cells arranged regularly . Model group: intimal thickening , intimal surface is not smooth , elastic fiber breakage , smooth muscle cell disorder , a lot of foam cell formation , showing large lipid infiltration plaque protruding into the lumen , inflammatory ceils proliferated. Xuezhikang : arterial wall structure clearer , intimal smooth, a small amount of foam -like changes in endothelial cells , smooth muscle cells arranged in a more regular , reduced proliferation of inflammatory cells . Qingzhi Tongmai particles low-dose group : Most of the clear structure , intimal smooth, occasionally foamy alter aortic endothelial ceils , smooth muscle cells arranged in a more regular , the proliferation of inflammatory cells decreased. Qingzhi Tongmai particles dose group : Active intimal smooth, occasionally a small amount of foamy slight changes in endothelial cells , smooth muscle ceils arranged in a more regular , inflammatory cells decreased. Qingzhi Tongmai particles high dose group : clear structure aortic intimal smooth, like changes in endothelial cell foam disappears , smooth muscle cells arranged rules , inflammatory cells decreased significantly. [ Conclusion ] Qingzhi Tongmai particles can reduce blood lipids, and its anti- atherosclerotic mechanisms may be related to lipid-lowering effects.
