摘要
目的研究象牙屑及其不同组分促伤口愈合的分子机制。方法象牙屑经脱钙、分级分离得3混合物,命名为MW14、MW5-14和MW5。通过2种动物创伤模型检验象牙屑对伤口愈合的功效;通过氯胺T法、明胶酶谱分析、qRT-PCR、ELISA和Western blotting等研究象牙屑及其不同组分促伤口愈合的机制。结果象牙屑可促进伤口愈合,其总水提物(WE)和MW5组分能显著激活PI3K/Akt和JNK/ERK信号通路,促成纤维细胞生长和胶原积累,提高MMP-2/-9活性和MMP-1/-13表达,抑制TIMP-1/-2表达,促进TGF-β、EGF、VEGF、CTGF、bFGF及TNF-α释放。结论象牙屑有效成分可能包括2部分:MW14和MW5;象牙屑促伤口愈合依赖于PI3K/Akt和JNK/ERK MAPK通路。
Objective To study the effect and mechanism of elephant tusk powder ( Elephas maximus L. ) on the wound healing. Methods Elephant tusk powder was demineralized, size-fractionated, and named as MW14, MW5-14 and MW5, respectively. A series of methods including MTT test, chloramine T and gelatin zymography assays, qRT-PCR, ELISA and Western blot were used to identify the effect of elephant tusk powder on the wound healing. Results Water extract ( WE ) of elephant tusk powder and MW5 significantly activated the PI3K/Akt and JNK/ERK pathways, promoted fibroblast growth and collagen production, elevated MMP2/9 activity and MMP1/13 expression, inhibited TIMP1/2 expression, as well as promoted the releases of TGF-β,EGF,VEGF,CTGF,FGF and TNF-α. Conclusion The main active constituents of elephant tusk powder may include two parts, namely MW14 and MW5. Elephant tusk powder promotes wound healing by activation of PI3K/Akt and JNK/ERK pathways.
