摘要
目的利用HPLC测定单面针茎中氯化两面针碱和橙皮苷的含量。方法采用Agilent Zorbax SB-C18柱(250 mm×4.6 mm,5μm),分别以0.02 mol/ml磷酸二氢钾:乙腈(74:26)、甲醇:水(40:60)为流动相,流速均为1.0 ml/min,柱温均为30℃,检测波长分别为328 nm和283 nm。结果氯化两面针碱,橙皮苷分别在0.072~0.36μg和0.1472~0.736μg范围内线性关系良好;平均回收率(n=5)分别为100.56%、100.86%。结论该方法为单面针药材的质量控制提供实验依据,该方法具有简便、快速、准确、重现性好的特点。
Objective The content of nitidin and hesperidin in the stem of zanthoxylum dissitum was determined by HPLC. Methods Agilent Zorbax SB-C18 (250mm × 4.6mm, 5μm)was used, a mobile phase of 0.02mol/ml KH2PO4- acetonitrile(74:26), methanol-water(40:60), the flow rate was 1.0 ml/min, the column temperature was 30℃, the detection was done at λ =328nm and λ =283nm. Results There was good linear relationship at the ranges of 0.072-0.36 μg and 0.1472-0.736 μg for nitidin and hesperidin. The average recoveries(n=5) were 100.56% and 100.86% respectively. Conclusion This method provides experimental basis for the quality control of single needle medicine and it is simple, rapid, accurate and good reproducibility.
