黄芪多糖对免疫性肝损伤小鼠协同刺激分子的影响研究 被引量:11

摘要 目的:观察黄芪多糖对免疫性肝损伤小鼠的肝功能和协同刺激分子及其受体的影响,探讨黄芪多糖治疗病毒性肝炎的作用机制,为其在临床上的应用提供理论依据。方法:小鼠随机分为4组:正常对照组、模型对照组、胸腺肽组、黄芪多糖组(0.2 g/kg),尾静脉注射卡介苗和脂多糖建立免疫性肝损伤小鼠模型。各组小鼠给予不同因素干预,造模结束后各组小鼠摘眼球采血,脱椎处死取肝脏。检测血清中丙氨酸氨基转移酶(ALT),流式细胞术检测脾脏中淋巴细胞表面CD28、CD80、ICOS、B7H2的表达,实时荧光定量聚合酶链反应(PCR)法检测脾组织中B7H3 mRNA含量。结果:黄芪多糖可降低血清中的ALT(P<0.05),模型对照组小鼠淋巴细胞表面的协同刺激分子表达均显著性增高(P<0.01或P<0.05)。黄芪多糖组CD28、CD80均显著降低(P<0.05),ICOS、B7H2无明显变化(P>0.05),B7H3 mRNA明显降低(P<0.01)。结论:黄芪多糖对免疫性肝损伤模型具有一定的保护肝脏和免疫调节作用,改善肝功能,可通过对部分协同刺激分子的影响调节免疫功能。 Objective : To observe the effect of astragalus polysaccharides (APS) on liver function, co - stimulatory molecules and their receptors of mice with immunological liver injury and to explore the therapeutic mechanism of APS in treating hepatitis and offering experimental evidence to its clinical application. Methods :32 rats were divided into 4 groups at random: normal group, model group, thymosin group, APS group(0.2 g/kg), with 8 rats in each group. Models of immunological liver injury were made by injecting bacillus calmette Guerin (BCG) via the vein of tail at the first day, and injecting lipopolysaccharide(LPS) at 1 lth day. After received different intervention factors, the mice were sacrificed, taking out liver and spleen and blood by excising the eyeballs. The serum was separated to determine alanine aminotrans- ferase (ALT). The levels of CD28, CD80,ICOS and B7H2 on surface of lymphocytes in spleen were detected by flow cy- tometry (FCM). B7H3 mRNA in spleen was detected by real -time polymerase chain reaction (PCR). Results:APS can reduce the levels of ALT in serum ( P 〈 0.05 ). In model group, the co - stimulatory molecules expressions of mice lym- phocyte surface were increased significantly (P 〈0.05 or 0.01 ). In APS group,CD28 ,CD80 and B7H3 mRNA were all significantly decreased ( P 〈 0.05 ) and ICOS and B7H2 had no significant changes ( P 〉 0.05 ). Conclusions : APS has certain protective and immunoregulative effects on immunological liver injury. It can improve liver function and regulate the immune function via the impact on parts of co - stimulatory molecules.
