摘要
目的:建立一个人类腭部涎腺腺样囊性癌细胞系并对其生物学特性作进一步探讨。方法:取人腭部小涎腺腺样囊性癌组织经原代培养并传代成系,命名为NACC细胞系。采用免疫组化法对NACC细胞系及另一涎腺腺样囊性癌细胞系SACC-83行多种细胞中间丝染色。结果:NACC细胞为亚三倍体核型、单层贴壁生长的上皮癌细胞系,异种接种可致瘤。NACC及SACC-83细胞均有抗keratin,GFAP,EMA,smoothmuscleactin,vimentin,S-100蛋白不同程度阳性。结论:NACC细胞系生长稳定,为源自干细胞的不同分化程度和发育周期的癌细胞所组成的细胞群体,可能含有肿瘤性闰管细胞、肌上皮细胞及未分化的多潜能干细胞等细胞成分,保持了腺样囊性癌的大部分特性。行体内、外试验时能较好地反映该类肿瘤的生物学特性。
Objective:To establish a salivary gland adenoid cystic carcinoma (ACC) cell line and analyze its biological behavior Methods:Primary culture method was used to establish a cell line named NACC from tumor blocks of a patient with palate ACC Biological behavior,cellular contents,chromosome of the cell line were analysed Immunohistiochemical stains were performed to identify seveal kinds of intermediate filaments on NACC Results:NACC is a monolayer anchorage dependence cells line with aneuploid triploidy Nude mice transplantation created tumor mass Positive stain of keratin,vimentin,smooth muscle actin,GFAP,EMA,Lys were demonstrated on NACC and SACC 83 cells respectively Conclusion:NACC cell line keep majority characteristic of ACC,could be a good symbol of ACC in vitro
