摘要
目的:探讨CD3AK细胞与LAK细胞的诱导、增殖动力学和杀伤活性的变化。方法:采用抗CD3单克隆抗体(anti-CD3McAb)和rIL-2及PHA(植物血凝素)共刺激法诱生扩增CD3AK细胞,用等量rIL-2和PHA共刺激法诱生LAK细胞,观察它们的增殖动力学变化;用MTT法以K562细胞和H7402细胞为靶细胞,测定CD3AK细胞和LAK细胞的杀伤活性。结果:微量的anti-CD3McAb辅以少量的rIL-2和PHA就能诱导和大量扩增CD3AK细胞,其扩增能力显著高于LAK细胞(P<0.001);CD3AK细胞对K562细胞和H7402细胞的杀伤活性显著高于LAK细胞(P<0.05)。结论:CD3AK细胞的扩增能力及杀伤活性均较LAK细胞为强。
Objective:To probe into the induction and changes of multiplication kinetics and killing activity in anti CD 3 monoclonal antibody activated killer cells (CD 3 AK) and lymphokine activated killer cells (LAK).Method:CD 3 AK cells were induced by combined anti CD 3 McAb and recombinant IL 2 (rIL 2),and PHA,and LAK cells were induced by the same dose of rIL 2 and PHA Their multiplication kinetics was observed.Killing activity of CD 3 AK and LAK cells was determined by MTT method with K 562 and H 7402 cells being used as target cells Results:CD 3,AK cells were stimulated by trace anti CD 3 McAb plus rIL 2 and PHA,and were multiplicated on a large scale The multiplication ability of CD 3 AK cells was higher than that of LAK cells markedly (P<0 001) and the killing activity of CD 3 AK cells was stronger than that of LAK cells significantly when K 562 cells and H 7402 cells were used as target cells (P<0 05) Conclusion:CD 3 AK cells had higher multiplication ability and killing activity in comparison with LAK cells CD 3 AK cell is another more efficient killer cell
